摘要
为了研究观赏南瓜离体快繁技术,以观赏‘金童'南瓜无菌苗为试验材料,茎尖与子叶为外植体,观察其在含有不同种类和浓度生长调节剂的培养基中的再生频率。试验结果表明,茎尖与子叶通过人工诱导培养均可以诱导出不定芽,在再生过程中,茎尖的芽诱导率比子叶高,子叶产生的愈伤组织明显高于茎尖;最佳芽诱导培养基组合为MS+6-BA 0.4 g·L^-1+IBA 0.05 mg·L^-1+30 g·L^-1蔗糖+7 g·L^-1琼脂。
In order to study the rapid reproduction in vitro technique of ornamental pumpkins, the 'Golden Kid (Jintong)' ornamental pumpkin aseptic seedlings were used as experimental materials, the shoot tips and cotyledons were used as explants, and their regeneration frequency in growth mediums which contained different kinds and concentrations of growth regulators were studied. The results showed that the adventitious buds could be induced by the shoot buds and cotyledons.. During the regeneration process, the bud induction rate of stem tips was higher than that of cotyledons, and the callus produced by cotyledons was significantly higher than the shoot tips;the best bud induction growth medium combination was MS+6-BA0.4 g·L^-1 +IBA0.05 mg·L^-1+ 30 g·L^-1 sucrose + 7 g·L^-1 agar.
