EMT相关调控因子Snail、Twist 1在口腔鳞状细胞癌中表达的相关性分析 被引量:2

摘要 目的研究上皮间充质转化(Epithelial-mesenchymal transition,EMT)的转录因子Snail、Twist 1与口腔鳞状细胞癌(OSCC)的表达及关系。方法实时定量PCR分析(RT-qPCR)技术和免疫组织化学(IHC)技术用于检测49例口腔鳞状细胞癌(OSCC)手术患者癌组织和癌旁组织中EMT相关转录因子SNAIL和TWIST 1的基因和蛋白的表达量。将二者的表达水平与临床病例信息相结合,分析OSCC患者发病因素,预防和治疗等方面与EMT之间的关系。结果 OSCC患者癌组织中Snail 蛋白及mRNA的相对表达量较癌旁组织高,Twist 1 蛋白和mRNA相对表达量较癌旁组织高(P<0.01)。Snail蛋白和mRNA的表达与T分期等无关(P>0.05),与吸烟、饮酒、淋巴结转移相关(P<0.05),Twist 1蛋白和mRNA的表达与患者性别、T分期、分化程度等无关(P>0.05),与淋巴结转移及吸烟情况相关(P<0.05)见表1-4。结论 Snail、Twist 1在OSCC中表达量较癌旁正常组织升高,并与转移及分化程度、吸烟、饮酒等因素相关,提示EMT相关调控转录因子Snail、Twist 1与OSCC的发生、发展及转移有着密切的联系。 Objective To investigate the expression and relationship of transcription factors Snail,Twist 1 and oral squamous cell carcinoma (OSCC) in epithelial-mesenchymal transition (EMT).METHODS: Real-time quantitative PCR analysis (RT-qPCR) and immunohistochemistry (IHC) were used to detect EMT transcription factors SNAIL and TWIST 1 in cancer tissues and adjacent tissues of 49 patients with oral squamous cell carcinoma (OSCC).The expression levels of protein and gene,combined with case information,analyzed the relationship between OSCC and EMT.Results The relative expression of Snail protein and mRNA in OSCC patients was higher than that in adjacent tissues.The relative expression of Twist 1 protein and mRNA was higher than that in adjacent tissues (P<0.01).The expression of Snail protein and mRNA was not associated with T stage (P>0.05),and was associated with smoking,drinking,and lymph node metastasis (P<0.05).The expression of Twist 1 protein and mRNA was not related to gender,T stage,differentiation degree,etc.(P>0.05),related to lymph node metastasis and smoking (P<0.05),see Table 1-4.Conclusion The expression of Snail and Twist 1 in OSCC is higher than that in adjacent normal tissues,and it is related to the degree of metastasis and differentiation,smoking,drinking and other factors,suggesting the occurrence and development of EMT-related regulatory transcription factors Snail,Twist 1 and OSCC.Transfer has a close connection.
