摘要
以红魔帝兜兰为试验材料,利用扫描电子显微镜对红魔帝花粉的超微结构进行观察,通过TTC(氯化三苯基四氮唑)染色对红魔帝兜兰的花粉活力进行研究,并用联苯胺-过氧化氢法测定其柱头可授性。结果表明,兜兰随着开放时间的延长花粉变硬,体积变小,花粉块黏性减弱;TTC染色法检测结果表明,花粉活力表现为开花10~30d>开花10d>花蕾期;柱头可授性测定结果表明,兜兰开花10~30d人工杂交成功率较高,其中10~15d授粉率最高。
Paphiopedilum maudiaewas used as the research material to observe the ultrastructure of P.maudiae type pollen by scanning electron microscope. The pollen viability of Paphiopedilum maudiae was studied by TTC(2,3,5-triphenyltetrazolium chloride) staining. The results showed that with the prolongation of the opening time, the pollen of a paphiopedilum became hard, the volume became smaller and the stickiness of the pollen mass decreased. TTC staining showed that pollen viability was > at 10-30 days after flowering and > 10 days after flowering. Stigma grafting assay showed that the success rate of artificial hybridization was a little higher within 10-30 days after flowering, and the pollination rate was the highest within 10-15 days after flowering.
