摘要
目的探讨槲皮素抗鼻咽癌(NPC)CNE2细胞的机制。方法通过CCK-8实验检测槲皮素在CNE2中的抗肿瘤活性;流式细胞术Annexin V-FITC//PI双染法检测细胞凋亡情况;采用Western blot检测槲皮素对CNE2细胞中凋亡标志蛋白、凋亡相关蛋白和凋亡诱导因子(AIF)以及细胞色素C(Cyt-C)的影响。结果槲皮素能抑制CNE2细胞生长、诱导细胞凋亡;Western blot结果显示,槲皮素能下调B淋巴细胞瘤/白血病-2蛋白(Bcl-2)和上调Bcl-2相关X蛋白(Bax)的表达;虽不能激活半胱氨酸天冬氨酸蛋白酶3(Caspase-3),但能通过切割DNA修复酶(PARP),促进AIF和Cyt-C从线粒体释放,从而诱导NPC细胞凋亡。结论槲皮素诱导鼻咽癌CNE2细胞发生了Caspase非依赖性细胞凋亡。
Objective To elucidate the anti-tumor activity of quercetin in nasopharyngeal carcinoma cell lines.Methods CCK-8 assay was employed to test the anti-tumor activity of quercetin. Cell apoptosis was detected by flow cytometry with AnnexinV-FITC/PI double-staining method. Apoptosis related protein was measured by western blot.Results Quercetin effectively inhibited cell proliferation in CNE2 cells by CCK-8 assay. Quercetin induced the cleavage of poly ADP-ribose polymerase(PARP) and triggered significant release of apoptosis inducing factor(AIF) and cytochrome C from mitochondria in a concentration-dependent and time-dependent manner.Conclusion Quercetin induced caspase-independent apoptosis in CNE2 Cells.
