依托咪酯联合富氢液对脓毒症小鼠氧化应激、炎症损伤情况及血清HMGB1、MDA水平的影响分析 被引量:5

摘要 目的观察依托咪酯联合富氢液对脓毒症小鼠氧化应激、炎症损伤情况及血清高迁移率族蛋白B1(high mobility group box 1,HMGB1)、丙二醛(malondialdehyde,MDA)水平的影响。方法选取雄性的Balb/c小鼠72只随机分成4组:正常对照组、脓毒症组、依托咪酯组、联合组,每组18只。首先以20 mg/kg的剂量腹腔注射脂多糖(lipopolysaccharide,LPS)进行脓毒症模型的建立,0.5 h后依托咪酯组以10 mg/kg腹腔注射依托咪酯、联合组以依托咪酯5 mL/kg+富氢液5mL/kg腹腔注射。比较LPS注射之后2、6 h的血清IL6及IL10浓度变化,6 h的肝肾及肺组织中HMGB1及MDA含量。结果与正常对照组相比,脓毒症组2、6 h的IL6均显著升高,与脓毒症组相比,依托咪酯组及联合组2、6 h的IL6均显著降低,与依托咪酯组相比,联合组2、6 h的IL6显著降低,差异均具有统计学意义, P 均<0.05;与正常对照组相比,脓毒症组2、6 h的IL10均显著降低,与脓毒症组相比,依托咪酯组及联合组2、6 h的IL10均显著升高,与依托咪酯组相比,联合组2、6 h的IL10显著升高,差异均具有统计学意义, P 均<0.05。与正常对照组比较,脓毒症组6 h肝肾肺组织中HMGB1、MDA含量明显升高,与脓毒症组比较,依托咪酯组及联合组6 h的HMGB1、MDA含量明显降低,并且联合组的HMGB1、MDA含量显著低于依托咪酯组,均有统计学差异,均 P <0.05。结论依托咪酯联合富氢液可减轻脓毒症小鼠的炎症损伤,并减少IL6、HMGB1和MDA的释放。 Objective To observe protective effects of the etomidate combined rich hydrogen fluid in the treatment of septic mice and on the expression of serum high mobility group box 1 (HMGB1),malondialdehyde(MDA). Methods Seventy-two male Balb/c mice were divided into four groups at random:normal control group,sepsis group,etomidate group,combined group,18 mice in each group.Lipopolysaccharide (LPS,20 mg/kg) was intraperitoneally injected to establish sepsis model,and then after 0.5 h,the etomidate group was intraperitoneally injected with etomidate at 10 mg/kg,and the combined group was intraperitoneally injected with etomidate 10 mg/kg + hydrogen-rich solution 5 mL/kg.At 2 h,6 h after LPS injection serum IL-6 and IL-10 were determined;At 6 h HMGB1 and MDA contents in liver,kidney and lung tissue were measured. Results Compared with the normal control group,IL-6 was significantly increased at 2 and 6 h in the sepsis group.Compared with the sepsis group,IL-6 was significantly decreased at 2 and 6 h in the etomidate group and the combined group.Compared with the normal control group,IL-10 was significantly decreased at 2 and 6 h in the sepsis group,and significantly increased at 2h and 6h in the etomidate group and the combination group ( P < 0.05 ). Compared with the normal control group,the HMGB1 and MDA contents in liver,kidney and lung tissues of the sepsis group were significantly increased at 6 h.Compared with the sepsis group,the HMGB1 and MDA contents in the etomidate group and the combined group were significantly decreased at 6 h.In addition,the HMGB1 and MDA contents in the combined group were significantly lower than those in the etomidate group ( P <0.05). Conclusion Etomidate combined with hydrogen-rich liquid can reduce inflammatory injury in septic mice and reduce the release of inflammatory factors,IL-6、HMGB1 and MDA.
