一测多评法同时测定蜘蛛果中3种成分 被引量:4

Simultaneous determination of three constituents in Campanumoea lancifolia by QAMS

摘要 目的建立一测多评法同时测定蜘蛛果中绿原酸、木犀草素、芹菜素的含有量。方法该药物甲醇提取液的分析采用Pntulips BP-C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-甲醇-0.1%磷酸水,梯度洗脱;柱温25℃;体积流量1.0 mL/min;检测波长340 nm。以绿原酸为内标,计算其他2种成分的相对校正因子,测定其含有量。结果绿原酸、木犀草素、芹菜素分别在0.022 00~0.351 3、0.006 312~0.101 0、0.000 527 4~0.041 20 mg/mL范围内线性关系良好(r≥0.999 8),平均加样回收率(RSD)分别为96.00%(1.5%)、 96.34(2.4%)、95.93(2.5%)。一测多评法所得结果接近于外标法。结论该方法稳定可靠,可用于蜘蛛果药材的质量控制。 AIM To establish a quantitative analysis of multi-components by single marker(QAMS) method for the simultaneous content determination of chlorogenic acid, luteolin and apigenin in Campanumoea lancifolia(Roxb.) Kurz.METHODS The analysis of methanol extract of this drug was performed on a 25 ℃ thermostatic Pntulips BP-C18 (4.6 mm×250 mm,5μm),with the mobile phase comprising of acetonitrile-methanol-0.1% phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 340 nm. With chlorogenic acid as an internal standard,the relative correction factors of luteolin and apigenin were established,followed by the determination of their contents.RESULTS Chlorogenic acid, luteolin and apigenin showed good linear relationships within the ranges of 0.022 00-0.351 3 mg/mL, 0.006 312-0.101 0 mg/mL, 0.000 527 4-0.041 20 mg/mL( r≥0.999 8),whose average recoveries(RSDs) were 96.00%(1.54%),96.34%(2.45%),95.93%(2.53%),respectively.The results obtained by QAMS approximated those obtained by external standard method.CONCLUSION The stable and reliable method can be used for the quality control of C. lancifolia.
