摘要 热休克蛋白(heat shock proteins,HSPs)是一类细胞含量最丰富、高度保守的分子伴侣,在所有的真核细胞中表达。细胞受到外界刺激,其合成增加,快速保护细胞对抗应激反应。热休克蛋白家族与肿瘤关系密切,HSP90调控肿瘤细胞增殖、分化和转移的多种相关蛋白活性,是肿瘤细胞形成抗药性/放射抗性的关键蛋白。抑制HSP90功能可推动下游活性蛋白降解,增加肿瘤细胞对治疗干预的易感性。本文综述了HSP90的生物特性和它在多种肿瘤发生发展中的作用及其靶向治疗等方面相关研究进展。 Heat shock proteins (HSPs) are a class of most abundant, highly conserved molecular chaperones, which expressed in all eukaryotic cells. When cells are stimulated by the stressful conditions, their synthesis increases, and they can quickly protect cells against stress. The HSPs family is closely related to tumors. HSP90 regulates the activity of various related proteins in tumor cells proliferation, differentiation and metastasis, and is a key protein for tumor cells formation resistance/radiation resistance. Inhibition of HSP90 function promotes degradation of downstream active proteins and increases susceptibility of tumor cells to therapeutic intervention. This article reviews the biological characteristics of HSP90 and its role in the development of various tumors and its targeted therapy.
