摘要
目的:研究高糖及高糖加胰岛素对雪旺细胞(SCs)培养的影响。方法:取2~4天的 SD大鼠的坐骨神经,培养和纯化SCs,将纯化的SCs分别置于普通培养液(对照组)、含10%的高糖培养液(观察组1)、含10%的高糖加4单位的胰岛素培养液(观察组2)进行SCs培养。结果:通过对培养SCs计数表明:观察1组SCs数量(29.800±3.668),对照组SCs数量(70.533±6.081)( t =-36.826, P =0.000);观察2组的SCs数量(68.533±5.449),观察1组SCs数量(29.800±3.668)( t =33.370, P =0.000);观察2组的SCs数量(68.533±5.449),对照组SCs数量(70.533±6.081)( t =-2.223, P =0.043)。结论:高糖环境明显不利于SCs的再生;而高糖加入胰岛素比高糖环境明显有利于SCs的再生,但高糖加胰岛素与普通培养基比较,也不太利于SCs再生,但差异无统计学意义。
Objective:To study the effects of high glucose and high glucose plus insulin on the culture of Schwann cells (SCs).Methods:The sciatic nerve of SD rats was taken for 2 to 4 days. The SCs were cultured and purified, and the purified SCs were separately placed in the normal culture solution (control group), containing 10 % high glucose medium (observation group 1), containing 10 % high sugar plus 4 unit of insulin culture solution (observation group 2).Results:The number of SCs in the observation groups 1 was 29.800±3.668, and the number of SCs in the control group was 70.533±6.081 ( t =-36.826, P =0.000);The number of SCs in the observation groups 2 was 68.533±5.449,and the number of SCs in the observation groups 1 was 29.800±3.668 ( t =33.370, P =0.000);The number of SCs in the observation groups 2 was 68.533±5.449, and the number of SCs in the control group was 70.533±6.081 ( t =-2.223, P =0.043).Conclusion:High glucose environment is not conducive to the regeneration of SCs. High glucose addition to insulin is more beneficial to the regeneration of SCs than high glucose environment. However, high glucose plus insulin is not conducive to SCs regeneration, but the difference is not significant.
