草还丹、人参固本丸和二至丸对D-半乳糖致衰老模型小鼠抗衰老作用的研究 被引量:6

摘要 目的探讨草还丹、人参固本丸、二至丸对D-半乳糖致衰老模型小鼠的抗衰老作用。方法 72只ICR小鼠,随机分为正常对照组、模型组、阳性对照组(VE,0.05g/kg)、草还丹组(4.5g/kg)、人参固本丸组(4.5g/kg)、二至丸组(4.5g/kg),每组12只,雌雄各半。除正常对照组外,其余各组采用颈背部皮下注射D-半乳糖(100mg/kg),建立小鼠衰老模型。造模同时,灌胃给予受试药,连续42天。观察小鼠体征,测量自主活动、学习记忆能力、抓力。末次给药后各组禁食不禁水,12h后摘眼球取血,对小鼠骨强度、血脂、睾丸系数、精囊腺系数进行测定。结果与模型组比较,三个复方组均能提高衰老小鼠抓力、骨强度、高密度脂蛋白胆固醇(HDL-C)、睾丸、精囊腺系数,降低Morris水迷宫持续时间和总路程、葡萄糖(GLU-OX)、甘油三酯(TG)、总胆固醇(TC)及低密度脂蛋白胆固醇(LDL-C)。草还丹组和人参固本丸组能提高小鼠自主活动。结论草还丹、人参固本丸和二至丸可延缓D-半乳糖所致衰老模型小鼠的衰老作用,改善精神萎靡、筋骨乏力、记忆减退、性腺萎缩等症状,该作用特征可认为是这三种中药复方延缓衰老的共同规律。 Objective To explore the anti-aging effect of Caohuandan, Ginseng guben pill and Erzhi pill on aging model mice induced by D-galactose. Methods A total of 72 healthy ICR mice were randomly divided into normal control group, model group, positive control group(VE, 0.05g/kg), Caohuandan group(4.5g/kg), Ginseng guben pill group(4.5g/kg), Erzhi pill group(4.5g/kg), 12 animals(half males and half females) in each group. Except for the normal control group, the other groups were subcutaneously injected with D-galactose(100mg/kg) at the back of neck to establish the aging model of mice. At the same time, the mice model was established by gavage for 42 consecutive days. The physical signs of mice were observed and the autonomous activities, learning and memory abilities and grasp were measured. After the last administration, each group was fasted to avoid water. After 12 hours, the eyeballs were taken and blood was collected. The bone strength, blood lipid, testis coefficient and seminal vesicle coefficient were determined. Results Compared to the model group, the grip strength, bone strength, high density lipoprotein cholesterol(HDL-C), testis and seminal vesicle gland coefficient in the three compound prescription groups increased, the duration and total distance of Morris water maze, glucose(GLU-OX), triglyceride(TG), total cholesterol(TC) and low density lipoprotein cholesterol(LDLC) decreased. The autonomous activity of mice in Caohuandan group and Ginseng guben pill group improved. Conclusion Caohuandan, Ginseng guben pills and Erzhi pills can delay the aging effect of D-galactose-induced aging model mice, improve the symptoms of mental depression, weakness of muscles and bones, memory loss, gonadal atrophy and so on, which can be regarded as the common law of these three traditional Chinese medicine compound prescriptions in delaying aging.
