摘要
基底细胞癌(basalcellcarcinoma,BCC)是皮肤科临床常见的非黑素瘤性皮肤癌,可侵犯深部组织,破坏邻近结构,易局部复发,较少发生远处转移。研究显示,Hedgehog信号通路的异常可导致BCC的发生。动物模型的建立进一步证实了该信号通路与BCC之间的密切联系,为研究疾病的治疗奠定了基础。Vismodegib等小分子抑制剂可阻断这一信号通路,抑制肿瘤生长。最近的基因组学研究指出,BCC的发展可能与额外的基因突变有关,这表明除了Hedgehog外,WNT、mTOR等其他信号通路在该肿瘤的发病机制中也具有重要意义。就Hedgehog信号通路对BCC发病的影响进行综述。
Basal cell carcinoma (BCC), a common non-melanoma skin cancer,is occasionally with deep invasion,destruction of adjacent structures,recurrence,but it rarely occurs distant metastasis.Some studies show that abnormal Hedgehog pathway may be associated with the occurrence of BCC.The establishment of animal models confirmed the close relationship between this pathway and BCC,which laid the foundation for the study of disease therapy.Small molecule inhibitors such as Vismodegib block this signaling pathway and inhibit tumor growth.Recent genomic studies point to the involvement of additional genetic mutations that might be associated with the development of BCC,suggesting significance of other signaling pathways,such as WNT,mTOR,aside from hedgehog in thepathogenesis of this human tumor,and this paper reviews the relationship between Hedgehog pathway and BCC.
