超声波协同纤维素酶提取黄精多糖的工艺优化研究 被引量:5

摘要 以黄精为原料、水为提取溶剂,采用超声波协同纤维素酶方法对其多糖提取。通过单因素试验和正交试验研究纤维素酶与底物质量比、超声时间、超声功率、浸提pH值、料液比对多糖提取率的影响。结果表明,影响最显著的是纤维素酶与底物质量比,其次是浸提pH值和超声功率,超声时间和超声功率的影响相对较小。优化得出最佳提取工艺为纤维素酶与底物质量比3%,超声功率100W,超声时间60min,料液比1∶8,浸提pH值5.0。与传统提取方法相比很大地提高了多糖提取率。 To use Polygonatum as raw material and water as solvent,synthetically of polysaccharides and sibiricoside was extracted from Polygonatum with cellulase enzyme under ultrasonic wave.The effect of the quality ratio of enzyme to substrate,ultrasonic power,ultrasonic time,material-liquid ratio,pH on the extraction of polysaccharides were investigated by single factor and orthogonal experiments.The results showed that the quality ratio of enzyme to substrate was the most significant factor affecting the synthetic extraction yield,the pH and ultrasonic power were the next place,and ultrasonic time,material- liquid ratio effects were the least.The optimum extraction conditions were as follows:the quality ratio of enzyme to substrate 3%,ultrasonic power 100 W,ultrasonic time 60 min,liquid-material ratio 1∶8,extraction pH 5.0.Compared with the traditional extraction method,the extraction rate of polysaccharide was greatly improved.
