摘要 目的:改进拟南芥叶片的SDS-聚丙烯酰胺凝胶电泳分离方法。方法:采用SDS-聚丙烯酰胺凝胶电泳法分离拟南芥叶片。主要比较和分析了两种样品溶解液和两种染色方法对实验的影响。改进的样品溶解液可防止样品的损失,节约样品和试剂;改进的染色方法操作时间短,灵敏度高,分离条带多且清晰。结论:改进的实验方法可以得到更为灵敏、清晰的实验结果,同时节约了实验经费和实验时间,减少了实验准备人员的工作量,更好地适应和促进了实验教学工作。 Objective:To improve the method of separating leaf proteins from Arabidopsis by Polyacrylamide Gel Electrophoresis(PAGE).Methods:Leaf proteins were separated by SDS-PAGE.The two different lysis buffers were used. Proteins were stained by primary Coomassie brilliant blue and stained by the blue silver,respectively.Results:The modified method could save the sample,exhibit clear protein bands,and shorten the period of staining-destaining process,which was an improvement of the traditional time-consuming method and could adapt to experiment teaching.
