血浆miRNA-200b及miRNA-21在浆液性卵巢癌中的表达特点分析

Expression Characteristics of Plasma miRNA-200b and miRNA-21 in Serous Ovarian Cancer

目的探讨血浆miRNA-200b及miRNA-21在浆液性卵巢癌中的表达特点。方法研究时间为2013年8月到2017年12月,采用回顾性、总结研究方法,选择浆液性卵巢癌患者60例作为卵巢癌组,同时选取同期患有良性上皮性卵巢肿瘤患者50例作为良性组,选择具有正常卵巢组织的40例患者作为对照组。采用荧光定量PCR检测3组患者血浆miRNA-200b及miRNA-21表达情况,调查患者的临床资料与其进行相关性分析。结果卵巢癌组中血浆miR-21、miRNA-200b相对表达量显著高于良性组与对照组(P <0. 05),miR-21、miRNA-200b在良性组与对照组中的相对表达量对比,差异无统计学意义(P> 0. 05)。不同临床分期、分化程度卵巢癌患者的miRNA-200b及miRNA-21表达量对比,差异都有统计学意义(P <0. 05)。在卵巢癌患者中,线性相关分析显示血浆miRNA-200b与miRNA-21表达量成正相关性(γ=0. 562,P <0. 05)。结论血浆miRNA-200b与miRNA-21在卵巢癌中呈现高表达状况,与患者的临床分期与组织学分化程度显著相关,两者可相互影响,在卵巢癌的发生、发展中发挥癌基因的功能。

Objective To investigate the expression characteristics of plasma miRNA-200 b and miRNA-21 in serous ovarian cancer. Methods 60 patients with serous ovarian cancer were selected as ovarian cancer group,and 50 patients with benign epithelial ovarian tumors were selected as benign group,and 40 cases with normal ovarian tissue were selected as the control group,the expression of plasma miRNA-200 b and miRNA-21 in the 3 groups of patients were detected by fluorescence quantitative PCR,and the clinical data and correlation analysis were investigated. Results The relative expression of plasma miR-21 and miRNA-200 b in the ovarian cancer group were significantly higher than that of the benign group and the control group( P <0. 05),and there were no significant difference in the relative expression of miR-21 and miRNA-200 b compared between the benign group and the control group( P > 0. 05). The expression of miRNA-200 b and miRNA-21 in ovarian cancer patients with different clinical stages and degree of differentiation were significantly different( P < 0. 05). In the ovarian cancer patients,linear correlation analysis showed that plasma miRNA-200 b were positively correlated with miRNA-21 expression(γ= 0. 562,P <0. 05). Conclusion There are high expression of plasma miRNA-200 b and miRNA-21 in ovarian cancer,and are highly correlated with the clinical stage and the degree of histological differentiation of the patients. Both of them can affect each other and play the function of the oncogene in the development of ovarian cancer.
