三根喷喉剂治疗慢性咽炎的机理研究

摘要 目的:探讨三根喷喉剂抗炎、镇痛及治疗慢性咽炎的作用机制。方法:采用小鼠毛细血管通透性实验,评价三根汤的抗炎作用;采用热板法、扭体法评价三根汤的镇痛作用;用氨水制备慢性咽炎大鼠模型,取咽喉组织HE染色,镜下观察三根汤对慢性咽炎大鼠咽喉的保护作用。结果:与对照组比较,盐酸小檗碱组,三根喷喉剂高剂量腹腔灌洗液A值降低(P <0. 01),三根喷喉剂中剂量组腹腔灌洗液A值有降低的趋势,但差异无统计学意义(P> 0. 05),低剂量组腹腔灌洗液A值无明显变化(P> 0. 05)。给药前各组小鼠痛阈值比较,差异无统计学意义(P> 0. 05);给药30 min后,与对照组比较,对乙酰氨基酚组、三根喷喉剂高剂量组的痛阈值显著性延长(P <0. 05),三根喷喉剂中剂量组、低剂量组的痛阈值有延长的趋势,但差异无统计学意义(P> 0. 05);给药60 min及给药90 min,各组间痛阈值均无显著性差异(P> 0. 05)。与对照组比较,对乙氨基酚组、三根喷喉剂高剂量组扭体次数显著性降低(P <0. 05),三根喷喉剂中剂量组、低剂量组的扭体次数有降低趋势,差异无统计学意义(P> 0. 05)。慢性咽炎模型大鼠咽部组织病理改变的实验结果显示,三根喷喉剂高、中、低剂量组均能显著改善氨水所致的大鼠咽喉炎症。结论:三根喷喉剂能降低慢性咽炎小鼠毛细血管通透性、延长小鼠痛阈值、降低小鼠扭体次数,改善咽炎小鼠咽喉的病理改变。 Objective: To observe the protective effect of Sangen Throat Spray( SGTS) on mice with chronic pharyngitis. Methods:The anti-inflammatory effect of SGTS was evaluated by capillary permeability test in mice. The analgesic effect of SGTS was evaluated by hot plate method and writhing method. The rat model of chronic pharyngitis was made by ammonia water,and HE staining of pharynx and larynx tissue was taken,and the protective effect of SGTS on rats with chronic pharyngitis under microscope. Results: Compared with the control group,the abdominal lavage fluid A values of berberine hydrochloride group and high-dose SGTS group decreased( P < 0. 01),while that of middle-dose SGTS group decreased,but there was no significant difference( P >0. 05). That of low-dose SGTS group did not change significantly( P > 0. 05). There was no significant difference in pain threshold between the all the groups before administration( P > 0. 05);30 minutes after administration,the pain thresholds of paracetamol group and high-dose SGTS group were significantly longer than that of the control group( P < 0. 05),while the pain thresholds of medium-dose SGTS group and low-dose SGTS group had a tendency to prolong,but there was no significant difference( P >0. 05). There was no significant difference in pain threshold between groups after 60 minutes’ and 90 minutes’ administration( P > 0. 05). Compared with the control group,the writhing times of the of ethaminophen group and the high-dose SGTS group decreased significantly( P < 0. 05),while the writhing times of the middle-dose SGTS group and the low-dose SGTS group decreased,but there was no significant difference( P > 0. 05). The experimental results of pathological changes of pharynx tissue in chronic pharyngitis model rats showed that the high-dose,middle-dose and low-dose groups of could significantly improve the inflammation of pharynx and larynx in rats induced by ammonia water. Conclusion: SGTS can reduce capillary permeability,prolong pain threshold,reduce writhing times and improve the pathological changes of throat in mice with chronic pharyngitis.
