Th22细胞及其细胞因子与不明原因复发性流产的关系研究 被引量:5

摘要 目的:探究辅助性T(Thelper,Th)细胞22细胞在不明原因复发性流产(unexplained recurrent spontaneous abortion,URSA)中的作用及其与Th17细胞之间的关系。方法:选择2016年10月至2018年5月在郑州大学第三附属医院妇产科门诊就诊的30例不明原因复发性流产患者为URSA组,选择同期在本院行人流术的30例正常早孕妇女为对照组。采用流式细胞仪检测两组孕妇外周血中Th22细胞和Th17细胞的数量,采用酶联免疫吸附试验检测两组孕妇血浆中白细胞介素(interleukin,IL)-22和IL-17A的浓度的变化。结果:与对照组相比较,URSA组患者外周血中Th22细胞的数量和Th17细胞的数量显著增多,血浆中IL-22和IL-17A的浓度同样显著升高;在URSA组中,Th22细胞数量与Th17细胞数量呈正相关,而在对照组中,未见明显相关性;在URSA组中,IL-22的浓度不仅与Th22细胞的数量呈正相关,也与Th17细胞的数量呈正相关,但与Th22细胞的关联强度较高。结论:Th22细胞的数量及其效应因子IL-22浓度的显著增高可能与URSA具有一定相关性,同时在URSA中,Th22细胞与Th17细胞呈相关,提示两种细胞的分化成熟,可能相互影响。 Objective:To investigate the role of Th22 cells in unexplained recurrent spontaneous abortion and its relationship with Th17 cells. Methods: Thirty women with unexplained recurrent spontaneous abortion in the Third Affiliated Hospital of Zhengzhou University from October 2016 to May 2018 were enrolled as URSA group. Thirty healthy pregnant women were selected as control group. Flow cytometry was used to detect the frequencies of Th22 cells and Th17 cells in the peripheral blood of the two groups. Enzyme-linked immunosorbent assay was used to detect the changes of IL-22 and IL-17A in the plasma of the two groups. Results: The percentages of Th22 and Th17 cells in peripheral blood of URSA patients were significantly increased than those in the control group, and the plasma concentrations of IL-22 and IL-17A of URSA patients were also significantly increased than those in the control group. There was a positive correlation between the Th22 and Th17 cells in the URSA group, but not in the control group. In the URSA group Positive correlation was found in the plasma concentration of IL-22 with Th22 cells, and also with Th17 cells. But the intensity of correlation with Th22 cells is relatively high. Conclusions: Increased frequencies of Th22 cells and high levels of IL-22 may be associated with USRA. And Th22 cells are correlated with Th17 cells, suggesting that the differentiation and maturation of the two cells may interact with each other.
