缺血再灌注损伤对H9C2心肌细胞活性的影响

摘要 目的为了探讨H9C2心肌细胞缺血低氧后恢复再灌注细胞活性与时间的关系,为后期研究内源性抗缺血再灌注(I/R)损伤的保护机制提供数据参考和实验前提。方法将相近代数的H9C2心肌细胞随机分为正常组(CON组)、缺血缺氧组(I组)(分别缺血缺氧4.0h,8.0h,12.0h,16.0h,20.0h)、I/R组(分别灌注0.5h,1.0h,1.5h,2.0h,3.0h,4.0h),使用CCK8试剂对各组的H9C2心肌细胞进行孵育,利用分光光度仪检测各组H9C2心肌细胞的活性。结果 I组H9C2心肌细胞与CON组比较,活性明显降低,差异具有统计学意义(n=4,P<0.05);I/R组的细胞活性在再灌注后1.0h内逐渐降低,在1.0h H9C2心肌细胞活性达到最低值,以后随时间的推移细胞活性逐渐增强(n=4,P<0.05)。结论缺血16.0h复灌1.0h时,H9C2心肌细胞缺血再灌注损伤最显著,为后期的实验模型的建立和分子保护机制研究提供了强有力的数据参考。 Objective To explore the relationship between the activity and duration of reperfusion of H9 C2 myocardial cells after hypoxia and reperfusion,and to provide data reference and experimental premise for the study of the protective mechanism of endogenous anti-ischemic reperfusion injury.Methods Similar algebraic H9 C2 cardiac muscle cells were randomly divided into normal group(CON group),ischemia hypoxia group(I group)(ischemia hypoxia respectively 4.0 h,8.0 h,12.0 h,16.0 h,20.0 h),ischemia reperfusion group(I/R group)(perfusion respectively 0.5 h,1.0 h,1.5 h,2.0 h,3.0 h,4.0 h),using CCK8 reagent H9 C2 cardiac muscle cells of each group incubation,using spectrophotometry to test the activity of each H9 C2 cardiac muscle cells.Results The activity of H9 C2 myocardial cells in the I group was significantly lower than that in the CON group,and the difference was statistically significant(n=4,P<0.05).The cellular activity of I/R group gradually decreased within 1.0 h after reperfusion,and the myocardial activity of H9 C2 reached the lowest value at 1.0 h,and gradually increased over time(n=4,P<0.05).Conclusion The experimental results showed that H9 C2 myocardial ischemia reperfusion injury was the most significant at 16.0 h after reperfusion for 1.0 h,providing a strong data reference for the establishment of the experimental model and the study of the molecular protective mechanism.
