一种中药复方制剂对急性髓系白血病细胞株MV4-11增殖和凋亡的影响

The effects of a chinese medicine regime on differentiation,proliferation and apoptosis of MV4-11 cell line

目的了解一种中药复方制剂对急性髓系白血病(AML)细胞株MV4-11增殖和凋亡的影响。方法用该中药复方制剂处理MV4-11细胞株,应用瑞氏染色方法观察细胞形态的变化,Cell Counting Kit-8法检测中药复方制剂对MV4-11细胞增殖的抑制情况,Annexin V法观察程序性坏死抑制剂与凋亡抑制剂处理MV4-11细胞株后其凋亡情况。结果该中药复方制剂对MV4-11细胞株的增殖起抑制作用(P<0.05),且呈剂量依赖关系,该中药复方制剂可诱导MV4-11细胞株的分化和凋亡。结论该中药复方制剂可能主要通过诱导白血病细胞株MV4-11分化和诱导凋亡的作用抑制白血病细胞的增殖。

Objective To study the effect of a Chinese medicine regime on the differentiation,proliferation and apoptosis of AML MV4-11 Cell Line.Methods MV4-11 cell line was cultured in vitro and treated with Chinese medicine regime.The cell proliferation of the leukemia cells was measured by CCK8 method.The differentiation of MV4-11 Cell Line was evaluated using Wright’s staining and the morphology was evaluated by light microscope.Annexin V assay was used to evaluate the apoptosis of MV4-11 cells after application of necrosis inhibitor and apoptotic inhibitor.Results The proliferation of MV4-11 cells treated with different concentrations of traditional Chinese medicine was inhibited(P<0.05) in a dose dependent manner and their differentiation was significantly increased.After treatment of the MV4-11 cell line with the Chinese medicine regime,apoptosis of the MV4-11 cells was significantly increased.Conclusion The Chinese medicine regime may inhibit the proliferation of AML MV4-11 cell lines mainly by inducing leukemia cell differentiation and inducing apoptosis.
