逐瘀散结颗粒抗大鼠子宫肌瘤作用机制研究

Study on the Mechanism of Zhuyu Sanjie Granule in the Treatment of Uterine Fibroid in Rats

目的观察逐瘀散结颗粒对子宫肌瘤大鼠性激素及细胞因子水平的影响,从而探讨其作用机制。方法SD大鼠随机分为空白组、模型组、桂枝茯苓丸组、逐瘀散结颗粒高、低剂量组(360、180mg·kg^-1),以苯甲酸雌二醇、黄体酮肌肉注射造大鼠子宫肌瘤模型,用放免法测定其雌二醇(E2)、孕酮(P)水平,ELISA检测血清IL-2、肿瘤坏死因子(TNF-α)含量,并进行子宫组织病理学检查。结果模型组大鼠表现出其子宫脏器系数增加,卵巢脏器系数减小;E2、P水平增加,血清IL-2水平下降、TNF-α水平升高;子宫平滑肌肌层增厚,排列紊乱,胞核增大。逐瘀散结颗粒高、低剂量组与模型组比较,子宫系数减小,卵巢系数增加,血清E2、P降低,血清IL-2水平升高,TNF-α水平降低。结论逐瘀散结颗粒可改善肌瘤子宫的病理学形态,降低血清E2、P,调节免疫功能,这些可能是其治疗子宫肌瘤的作用机制。

Objective To observe the effect of Zhuyu Sanjie Granule on the levels of sex hormones and cytokines in uterine fibroid rats, and to explore its mechanism of action. Methods SD rats were randomly divided into blank group, model group, Guizhi Fuling group, Zhuyu Sanjie Granule high and low dose group (360, 180mg· kg^- 1 ). Rat model of uterine fibroids was induced by intramuscular injection of estradiol benzoate and progesterone. The levels of estradiol (E2) and progesterone ( P) were measured by radioimmunoassay. Serum IL-2 and tumor necrosis factor content (TNF -α) were detected by ELISA , and uterine histopathological examination was performed. Results The rats in the model group showed an increase in uterine organ coefficient and a decrease in ovarian organ coefficient;E2 and P levels increased ,serum IL-2 levels decreased, and TNF -α levels increased;uterine smooth muscle layer thickened and arranged disorderly The nucleus is enlarged. Compared with the model group, the uterine coefficient decreased, the ovarian coefficient increased, the serum E2 and P decreased, the serum IL-2 level increased and the TNF - alpha level decreased in the high and low dose groups of Zhuyu Sanjie Granule. Conclusion Zhuyu Sanjie Granule can improve the pathological morphology of uterine fibroids, reduce serum E2 and P, and regulate immune function. These may be the mechanism of its treatment of uterine fibroids.
