摘要
目的评价他扎罗汀凝胶治疗深部组织损伤的效果,观察创面中血管内皮生长因子(VEGF-α)的表达水平。方法采用磁铁压迫法建立小鼠深部组织损伤模型后,将小鼠随机分为3组,模型组不做任何处理,他扎罗汀组于深部组织损伤部位sc他扎罗汀凝胶0.2 mg,康惠尔组sc康惠尔凝胶0.2 mg,每隔48 h注药1次。拍照测量第1、3、5、7、10、12、14天治疗后的创面愈合面积比;第14天创面取材,HE染色观察创面愈合过程中组织结构改变;Western blotting、RT-PCR法检测组织中VEGF-α的蛋白和基因表达水平。结果建模后第10、12、14天,康惠尔组和他扎罗汀组的创面愈合比均显著低于模型组,两组比较差异有统计学意义(P<0.05)。HE染色显示,与模型组相比,他扎罗汀组成纤维细胞较多,新生毛细血管形成,与阳性对照康惠尔组结果基本一致;Western blotting和RT-PCR结果显示他扎罗汀组中VEGF-α蛋白和基因表达水平显著高于模型组,两组比较差异有统计学意义(P<0.05)。结论他扎罗汀凝胶能促进小鼠深部组织损伤的愈合,提高创面中VEGF-α的表达。
Objective To evaluate the effect of Tazarotene Gel on deep tissue injuries, and to observe the expression level of vascular endothelial growth factor(VEGF-α) in wounds. Methods After establishing a deep tissue injury model in mice by magnet compression, the mice were randomly divided into three groups. The model group did not receive any treatment. The tazarotene group was injected subcutaneously with the commercial Tazarotene Gel 0.2 mg at the deep tissue injury site, and Purilon Gel group was given a subcutaneous injection of the purilon gel 0.2 mg as a positive control. Drugs were injected every 48 h. The wound healing ratio was measured on the 1 st, 3 rd, 5 th, 7 th, 10 th, 12 th and 14 th day after the treatment, and the changes of tissue structure was observed on the 14 th day by HE staining. Western blotting and RT-PCR were used to detect the protein and gene expression levels of VEGF-α in tissues. Results After modeling for 10, 12, and14 d, the wound healing ratio of purilon and tazarotene groups were significantly lower than those in the model group, and there were significant difference between two groups(P < 0.05). HE staining showed that compared with the model group, tazarotene group had more fibroblasts and new capillary formation, which was consistent with the positive control of purilon gel group. Western blotting and RT-PCR results showed that the expression of VEGF-α protein and gene in the tazarotene group was significantly higher than that in the model group, with significant difference between two groups(P < 0.05). Conclusion Tazarotene Gel can promote the healing of deep tissue injuries in mice and increase the expression of VEGF-α in wounds.
