老鹳草素对去势抵抗性前列腺癌细胞增殖与迁移能力的影响 被引量:2

摘要 目的探讨老鹳草素对去势抵抗性前列腺癌细胞增殖与迁移能力的影响及机制。方法培养人前列腺癌DU145 细胞株,分别用浓度为0、30、60、100 滋mol/L 的老鹳草素培养液作用于DU145 细胞。MTS 法检测老鹳草素对DU145 细胞增殖的影响;划痕实验和Transwell检测细胞的迁移能力;Western-blot 检测细胞增殖及迁移相关蛋白的表达水平。结果MTS 显示老鹳草素呈浓度依赖性抑制DU145 细胞增殖(P< 0.05);划痕实验和Transwell显示老鹳草素能够显著抑制DU145 细胞的迁移能力(P< 0.05);经老鹳草素干预后,c-Myc蛋白表达水平较对照组显著降低,E-cadherin 蛋白表达水平较对照组显著增加。结论老鹳草素可显著抑制去势抵抗性前列腺癌细胞DU145 的增殖和迁移能力。 Objective To explore the effects and mechanism of Geraniin on the proliferation and migration of castration-resistant prostate cancer cells. Methods DU145 cells were cultured with 0,30,60 and 100 umol/L Geraniin,respectively.MTS assay was performed to detect the inhibition of DU145 cell proliferation by Geraniin at different concentrations;Scratch test and Transwell chamber test were performed to detect cell migration ability;Western blot was performed to detect the c-Myc protein and E-cadherin protein expressions. Results MTS results showed that Geraniin could significantly inhibit the proliferation of DU145 cells(P< 0.05). Scratch test and Transwell test showed that Geraniin could significantly inhibit the migration of DU145 cells compared with the control group (P< 0.05). After intervention with Geraniin, the expression of c-Myc protein decreased significantly, while the expression of E-cadherin protein increased significantly. Conclusion Geraniin could significantly inhibit the proliferation of castration-resistant prostate cancer cell line DU145 and inhibit its migration ability.
