腹部开放性海水浸泡伤BALB/c小鼠模型的建立 被引量:7

Mouse model for open wounds of abdomen with seawater immersion injury

摘要 目的建立腹部开放性海水浸泡伤BALB/c小鼠模型。方法选取健康雄性BALB/c小鼠62只,随机分为3组:模型对照组(A组,经腹腔开放伤预处理)、海水浸泡组(经海水浸泡60 min二次打击,B1组予以25℃海水浸泡,B2组予以20℃海水浸泡)。于建模处置后0、2、4、6 h对3组小鼠进行肛温检测,并观察海水浸泡伤后的存活率;于建模处置后6、12、24、48 h观察小鼠的肺、肠壁和肝组织病理学变化。分析不同温度对腹部开放性海水浸泡伤BALB/c小鼠存活率的影响,观察小鼠肺、肠壁、肝组织病理、肛温、存活率等指标,探索建立腹部开放性海水浸泡伤BALB/c小鼠模型。结果 20℃海水浸泡组小鼠2 h全部死亡,25℃海水浸泡组12 h后存活率80%;与对照组比较,腹腔开放伤预处理、海水浸泡二次打击后的小鼠肛温明显降低,各脏器(肺、肠、肝)等均呈现不同程度的炎症反应。结论小鼠经腹腔开放伤预处理,25℃海水浸泡二次打击后的模型,存活率高,肛温及各脏器病理变化均符合海水浸泡伤特点。该研究成功建立了开放性海水浸泡伤BALB/c小鼠模型。 Objective To establish a mouse model for open wounds of the abdomen with seawater immersion injury.Methods Sixty-two male BALB/c mice were randomly assigned to three groups:the control group(group A,only the open wound of abdomen was treated)and the two seawater immersion groups(the open wound of abdomen model was pretreated before being immersed in seawater for 60 minutes,group B1 immersed in seawater at 25℃,and group B2 immersed in seawater at 20℃). Rectal temperature was monitored at 0,2,4 and 6 h after operation,and survival rate was observed respectively. The pathological changes in the liver,lung and intestines were observed at 6,12,24 and 48 h after operation.The pathological changes in organs,rectal temperatures and survival rates were studied respectively. Results All the mice in the seawater immersion group at 20℃ died in 2 hours,and the survival rate was 80% after 12 hours in the seawater immersion group at 25℃. Compared with the control group,mice challenged with both the open wound of the abdomen and seawater immersion showed lower rectal temperatures,but more typical pathological changes associated with inflammation. Conclusion Mice treated with 25℃ seawater show a proper survival rate,lower rectal temperatures and pathological changes consistent with the characteristics of seawater immersion injury,suggesting that we have successfully established a mouse model for open wounds of the abdomen with seawater immersion injury.
