基于转录组测序的花生籽粒不同发育时期油脂合成相关基因差异表达分析 被引量:5

Differential Expression Analysis of Genes Related to Lipid Synthesis through Transcriptome Sequencing during Different Developmental Stages in Peanut Seed

摘要 为研究花生籽粒不同发育时期油脂合成过程中基因的表达调控模式,对花后20、35、50、60d的花生籽粒(分别表示为HS20、HS35、HS50、HS60)进行转录组测序。通过对转录组测序数据进行分析,HS20、HS35、HS50、HS60样品的文库分别获得了109321068、106867224、109313082、107123540条Cleanreads。将得到的花生籽粒转录组数据按发育时期的先后顺序进行两两比对,HS20-vs-HS35、HS35-vs-HS50和HS50-vs-HS60的差异表达基因总数分别是25769、18403、12308个,其中上调表达基因分别占12.55%、62.92%、26.28%。对差异表达基因进行KEGGPathway分析,得到135条代谢通路,其中与油脂合成相关的有脂肪酸生物合成、不饱和脂肪酸生物合成、脂肪酸延长、脂肪酸代谢、花生四烯酸代谢等代谢通路。 In order to survey the regulation patterns of genes expression in oil synthesis during the different developmental stages in peanut seed,the peanut seeds of20,35,50,60 days after flower(represented as HS20,HS30,HS50,HS60 respectively) were sequenced for transcriptome.Through the analysis of transcriptome sequencing data,the clean reads of HS20,HS35,HS50,HS60 sample library were109 321 068,106 867224,109 313 082,107123 540 fragments respectively.The transcriptome data were pair-wise compared in period order,and the total quantities of differential expression genes of HS20 -vs-HS35,HS35-vs-HS50 and HS50-vs-HS60 were25 769,18 403 and12 308 respectively,of which up-regulated genes accounted for12.55%,62.92% and26.28% respectively.KEGG Pathway analysis showed that,all differential expression genes were classified into135 KEGG pathways,of which fatty acid biosynthesis,biosynthesis of unsaturated fatty acids,fatty acid elongation,fatty acid metabolism and arachidonic acid metabolism KEGG pathways were related to lipid synthesis.
