热疗联合旋毛虫肌幼虫排泄分泌蛋白对小细胞肺癌H446增殖的影响 被引量:2

摘要 目的:观察热疗联合旋毛虫肌幼虫排泄分泌蛋白(ESPs)对小细胞肺癌H446细胞增殖有无抑制作用。方法:将对数生长期的H446细胞随机分为4组。对照组:H446细胞培养过程中不作任何处理;热处理组:将细胞消化后放置离心管中在42℃水浴中加热2h;排泄分泌蛋白处理组:将提取的旋毛虫肌幼虫排泄分泌蛋白用无血清培养基稀释成0.15mg/ml、0.3mg/ml、0.6mg/ml,分别与H446细胞共培养;热处理联合排泄分泌蛋白组:先经水浴处理后加入各浓度的ESPs。采用噻唑蓝比色法(MTT法)检测细胞抑制率。结果:MTT检测结果显示,随着ESPs浓度的增加及作用于H446细胞时间的延长,H446细胞的增殖活力逐渐减弱,热疗联合ESPs对H446细胞具有明显的抑制作用,呈剂量—时间依赖性效应。结论:42℃加热2h联合旋毛虫肌幼虫排泄分泌蛋白对小细胞肺癌H446细胞的增殖有抑制作用。 Objective:To observe the effect of hyperthermia and Trichinella spiralis muscle-larva excretory-secretory proteins(ESPs) on proliferation rate of H446 lung cancer cells.Methods:Exponentially growing H446 cells were divided into control group,hyperthermia treatment group,ESPs treatment group and combined hyperthermia and ESPs therapy group.After the digestion,the cells in hyperthermia treatment group were placed the centrifuge tube hated in 42℃ water bath for 2 hours;the cells in ESPs therapy group with 0.15mg/ml,0.3mg/ml,0.60mg/ml ESPs;the cell in combined hyperthermia and ESPs therapy group were placed water bath before ESPs,the cell in control group were cultured without special treatment.The effects of hyperthermia and ESPs on the proliferation activity of H446 cells were detected by MTT colorimetry.Results:The results of MTT detection showed that the proliferation of H446 cells decreased with the increase of the concentration of ESPs and the prolongation of the time of H446 cells,and hyperthermia and ESPs had a significant inhibitory effect on H446 cells,showing a dose time effect.Conclusion:Hyperthermia at 42℃ for 2 hours combined with ESPs could inhibit cell proliferation.
