摘要
小分子热激蛋白(small heat shock proteins, sHSP),作为一种分子伴侣,在植物逆境胁迫响应中发挥着重要的作用。本研究克隆了水稻中2个编码小分子热激蛋白的基因 Os16.9A和 Os16.9B, Os16.9A和 Os16.9B编码的150个氨基酸,相似性高达99%。 Os16.9A和 Os16.9B的转录方向相反,2个基因的起始密码之间的距离为2.6 kb,它们的启动子均含有CAAT-box、TATA-box、GATA-box以及与热激等胁迫应答有关的顺式作用元件HSE和ACGT aterd1;Os16.9A的启动子还包含响应冷、干旱应答的顺式作用元件LTRE,这些顺式作用元件呈不对称分布,推测这2个基因间的序列具有双向启动子的功能,且这2个基因可能受胁迫诱导表达。该研究将为进一步研究水稻 Os16.9A和 Os16.9B基因的表达调控及其功能提供参考。
As a molecular chaperone, small heat shock proteins(sHSPs)plays an important role in plant stress response. In this study, two small molecular heat shock protein genes in rice,named Os16.9A and Os16.9B were cloned,they both encoded 150 amino acids, with 99% similarity.The transcriptional direction of Os16.9Aand Os16.9B is opposite and the distance between the initial codon of the two genes was 2.6 kb.The results of promoter element analysis show that both the promoters of these two genes contained CAAT-box, TATA-box, GATA-box, as well as the cis-acting elements HSE and ACGT aterd1 which responded to stress such as heat shock.And ACGT aterd1 and so on. The promoter of Os16.9A also contains the cis-acting element LTRE, which responds to cold and drought. These cis-acting elements are asymmetrically distributed in the promoters of Os16.9A and Os16.9B. So it is reasonable to assume that the sequence between the two genes may have the function of bidirectional promoter and the two genes may express under stress conditions and in a particular period of plant growth. Cloning and sequence analysis of promoters of Os16.9A and Os16.9Bgenes will provide a reference for further study on expression regulation and functional analysis of Os16.9A and Os16.9B genes in rice.
