摘要
目的:建立食品中鸡、鸭源性成分的单向测序鉴定方法。方法:动物组织经蛋白酶K消解后,用全自动核酸提取仪提取DNA,以提取DNA作为模板,由特异引物进行PCR扩增,扩增产物经毛细管电泳仪鉴定,将目的扩增条带回收后进行测序鉴定。结果:鸡、鸭两种动物的目的扩增条带经单向测序后分别与SN/T2978-2011及SN/T3731.5-2013中鸡、鸭参考序列进行同源比对,同源性达到100%。本方法的鸡、鸭肉最低检出限为100mg·kg^-1。结论:该方法简单、准确、成本低,适用于食品中鸡、鸭源性成分鉴定。
Objective: To establish a one-way sequencing method for identification of chicken or duck-derived materials in food products. Methods: Animal tissues were digested with protease K and extracted with a fully automated Nucleic Acid Extraction System. Extracted DNA was used as a template and amplified by specific primers. The amplified products were identified by capillary electrophoresis. The objective amplified bands were extracted by Gel Electrophoresis and identified by sequencing. Results: After one-way sequencing,the objective amplified bands of chicken and duck were homologously aligned with the reference sequences of chicken and duck in Entry-exit criteria of SN/T2978-2011 and SN/T 3731.5-2013. The sequence homology reached one hundred percent. The minimum detection limit of this method is 100 mg·kg^-1. Conclusion: This method is simple,accurate and low cost,which is particular suitable for the identification of chicken or duck-derived materials in food products.
