新会陈皮提取物中四种黄酮成分含量测定及其抗炎活性研究 被引量:15

摘要 目的:比较不同陈皮成分之间的生物活性差异,并分析提取物中不同黄酮成分含量与抗炎活性之间的关联。从成分与活性方面为“陈皮,陈久者良”寻找科学依据。方法:采用HPLC法测定11种不同储存年限的新会陈皮的提取物中4种黄酮成分的含量,用1μg·mL^-1的脂多糖刺激小鼠RAW264.7巨噬细胞,构建炎症模型,测定提取物的体外抗炎活性。结果:在11种新会陈皮样品中均检测到4种黄酮化合物,随着存储时间的增加,提取物中橙皮苷的含量有增加趋势,11个样品中,抑制RAW264.7巨噬细胞产生NO的IC50值均小于50μg·mL^-1,提取物中川陈皮素的含量与抗炎活性的关联系数为0.815。结论:存储时间越久、橙皮苷含量越高的趋势可为“陈皮,陈久者良”提供理论支撑,11种新会陈皮的提取物均具有良好的抗炎活性,其抗炎活性的强弱与川陈皮素的含量有一定的相关性。 Objective: The biological activities of different components of citrus reticulata ‘Chachi’ were compared and the correlation between different flavonoid content and anti-inflammatory activity was analyzed. From the aspects of composition and activity found scientific basis for “the older,the better”. Methods: HPLC was used to analyze the content of four flavonoids in the ethyl acetate extracts of Citrus reticulata ‘Chachi’. In vitro inflammation model of LPS-stimulated RAW 264.7 cells was used to determine the anti-inflammatory effect of ethyl acetate extracts. The correlation between the flavonoids content and biological activity was analyzed. Results: Four flavonoid compounds in extracts of 11 samples were detected,the anti-inflammatory effect of these samples showed significant dose-response relationship and the IC50 values of 11 samples that inhibited NO production were less than 50 μg·mL^-1. The correlation coefficient between nobiletin and anti-inflammatory activity was 0.815. Conclusion: The content of hesperidin in the extract increased with the increase of storage time that can provide theoretical support for “the older,the better”. 11 ethyl acetate extracts have good antiinflammatory activity and the anti - inflammatory activity was correlated with the content of nobiletin.
