摘要
目的探究脑缺血再灌注对小鼠学习记忆及Bax、Bcl-2表达的影响。方法选用健康的4周龄小鼠20只,随机分为空白组10只,实验组10只:实验组小鼠夹闭双侧颈总动脉,脑缺血20min后复灌,建立缺血再灌注模型,空白组小鼠不做任何处理;实验组和空白组小鼠于造模后3天用Morris水迷宫进行检测,观察每组小鼠术后空间学习行为学的变化;采用免疫组织化学法,对比两组小鼠脑中海马区Bax、 Bcl-2蛋白表达差异。结果水迷宫实验中实验组小鼠逃避潜伏期与空白组小鼠相比时间延长,差异有统计学意义(P<0.05)。脑片免疫组化结果显示实验组小鼠海马区第9天Bax、Bcl-2阳性细胞表达量与空白组相比增多,差异有统计学意义(P<0.05)。结论提示脑缺血再灌注使小鼠学习记忆能力下降,可能与海马区Bax、Bcl-2的表达增多有关。
Objective To investigate the effect of Bax and Bcl-2 protein in brain ischemia-reperfusion injury in mice. Methods The models of this experiment were made of 20 Kunming mice aged 4 weeks. They were randomly divided into normal control group and ischemia-reperfusion model group. Both sides of common carotid arteries of the last group were clipped and then recanalized after 20 minutes, and do nothing to the normal control group. After 3 days of models onset, the two groups were tested by Morris water maze to observe the changes of spatial learning behavior of them after surgery. After that, the differences in expression of Bax and Bcl-2 proteins in hippocampus with immunohistochemisty between the two groups were contracting. Results From the Morris water maze experiment, the ischemia-reperfusion had longer escape latency than the common control one. There was statistical difference between two groups(P<0.05). The results of the immunohistochemisty showed the expression of Bax and Bcl-2 in the hippocampus of the ischemia-reperfusion group increased after 9 days compared with the common control group. Conclusion Ischemia reperfusion decreased the learning and memory ability of mice, which may be related to the increased expression of Bax and.Bcl-2 in the hoppocampus.
