桂皮醛对高糖环境下MC3T3-E1成骨细胞增殖与分化的影响 被引量:6

摘要 目的:观察桂皮醛对高糖作用下MC3T3-E1成骨细胞增殖与分化的影响。方法:用0(对照组)、10、20、40mg/L的桂皮醛分别在葡萄糖(25mmol/L)的环境下培养MC3T3-E1细胞,培养1、4、7d后检测细胞增殖、碱性磷酸酶活性,茜素红染色观察钙化结节,鬼笔环肽染色观察细胞结构框架以及成骨相关基因骨钙蛋白(osteocalcin,OCN)mRNA表达水平。结果:桂皮醛可促进高糖环境下成骨细胞增殖及ALP活性(P<0.05);矿化结节数量增多且体积增大;实验组OCNmRNA表达水平明显上调(P<0.05);细胞骨架染色结果示:当桂皮醛浓度为10、20mg/L时细胞铺展面积增大且细胞骨架更加清晰。结论:桂皮醛可促进高糖作用下MC3T3-E1成骨细胞的增殖与分化。 Objective: To observe the effects of cinnamaldehyde (CA) on the proliferation and differentiation of MC3T3-E1 osteoblasts induced by high glucose.Methods: MC3T3-E1 cells were cultured in glucose (25 mmol/L) with 0 (control),10,20,and 40 mg/L CA.Cell proliferation,ALP activity,and alizarin red staining was measured.Morphology of the cytoskeleton was observed by confocal laser scanning microscope.Expression levels of osteogenesis related genes OCN mRNA were detected by real-time PCR.Results: CA could promote the proliferation and ALP activity of osteoblasts in high glucose environment ( P <0.05).The number and the volume of mineralized nodules increased.The expression of OCN mRNA in the experimental group was significantly up-regulated ( P < 0.05 ).When the concentration of CA was 10mg/L and 20mg/L,the cell spreading area was increased and the cytoskeleton was clearer.Conclusion: CA can promote the proliferation and differentiation of MC3T3-E1.
