摘要
目的:探究穿心莲内酯(Andro)对镉致小鼠急性肝损伤的改善作用。方法:选择30只ICR小鼠,随机分成空白组、模型组、Andro低剂量组和Andro高剂量组,其中空白组6只,其余组各8只。将空白组和模型组按每10g体重0.1mL灌胃0.5%羧甲基纤维素钠,Andro低、高剂量组分别按每10g体重灌胃0.1mL30、60mg/kg的Andro,连续灌胃7d,期间小鼠自由采食和饮水。第7天灌胃2h后对模型组和Andro给药组腹腔注射4mg/kgCdCl2,18h后脱臼处死小鼠,分离血清测定丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)及肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、IL-1β含量。分离肝脏,测定肝组织中还原性谷胱甘肽(glutathione,GSH)和超氧化物歧化酶(SOD)含量,HE染色检测肝脏组织病理学变化。结果:CdCl2(4mg/kg)可显著升高小鼠血清中ALT、AST酶活力(P<0.01),显著升高小鼠血清中TNF-α、IL-1β与IL-6的含量(P<0.01),同时显著降低肝脏中GSH的含量与SOD的活力(P<0.01);给予30mg/kg或60mg/kg的Andro能显著降低血清ALT、AST活力和TNF-α、IL-1β、IL-6含量(P<0.01),提高肝脏中GSH含量和SOD活力(P<0.05)。结论:Andro对镉致急性肝损伤具有一定的改善作用。
Objective:To investigate the amelioration of andrographolide(Andro)on cadmium-induced acute liver injury in mice.Methods:Thirty ICR mice were randomly divided into four groups:blank group,model group,low-dose Andro group and high-dose Andro group.The blank group and model group were given 0.5%sodium carboxymethyl cellulose(0.1 mL/10 g.b.w),the other groups were treated with Andro(30,60 mg/kg)for 7 days.On the 7th day,the model group and Andro group were intraperitoneally injected with 4 mg/kg CdCl 2 in 2 hours later of last administration.After 18 hours,the mice were executed.Serum levels of ALT,AST,TNF-α,IL-1β,IL-6 were measured.The liver was separated and the contents of GSH and SOD in liver tissue were measured.The pathological changes of liver tissue were detected by H&E staining.Results:Andro could reduce the activity of ALT,AST,TNF-α,IL-1βand IL-6 in serum and enhance the activity of SOD and GSH in liver,which had statistical significance compared with model group(P<0.01,P<0.05).Conclusion:Andro has certain amelioration on cadmium-induced acute liver injury.
