基于实体语法系统研究中药治疗乳腺癌的分子调控网络 被引量:3

摘要 目的:乳腺癌是常见的女性恶性肿瘤之一,其发病机制复杂,而中药多成分、多靶点的特点使其在治疗乳腺癌的过程中的具体作用也更加难以明确,因此本文系统地对中药治疗乳腺癌的作用机制进行研究。方法:本研究基于构建的乳腺癌分子调控网络,利用实体语法系统分析常用治疗乳腺癌的中药对乳腺癌相关靶点的作用,从而对其治疗涉及的分子机制进行解析。结果:通过多个数据库收集及文献验证得到包含213个节点以及486条边的乳腺癌分子调控网络。利用筛选的4味中药通过实体语法系统进行作用机制解析,最终表明所选择的中药中有48个化学成分会对ESR1、AR、BCL2、PGR、ERBB2、SRC、PIK3CA、CA2、EGFR、HSP90AA1、PIK3CG、KDR、INSR、TFF1、PLAU 15个乳腺癌相关靶点进行调节,影响多种生物过程从而达到治疗疾病的目的。结论:本研究从分子调控网络方向系统的解析了中药治疗乳腺癌的作用机制,为新药开发及有效成分配伍提供了参考。 Objective: Breast cancer is one of the common female malignant tumor, its pathogenesis is complex and has not been clearly studied. Chinese medicines have characteristics of multiple ingredients, multiple targets, which makes it more difficult to determine the specific role of the drug in the treatment of breast cancer, so we decide to systematically study the mechanism of traditional Chinese medicine (TCM) for treatment of breast cancer. Methods: Based on the established molecular regulatory network of breast cancer, this study analyzed the effects of TCM, which is commonly used to treat breast cancer, on the key genes of breast cancer by using the entity grammar system, so as to analyze the molecular mechanism involved in its treatment. The molecular regulatory network of breast cancer was obtained through data collection and literature validation. The network contained 213 nodes and 486 edges. The analysis of the mechanism of action by using the selected TCM through the entity grammar system finally indicates that 48 chemical components of the selected TCM would regulate ESR1, AR, BCL2, PGR, ERBB2, SRC, PIK3CA, CA2, EGFR, HSP90AA1, PIK3CG, KDR, INSR, TFF1, PLAU 15 breast cancer-related targets and regulate a variety of biological processes so as to achieve the purpose of treating diseases. Conclusion: This study systematically analyzed the mechanism of TCM in the treatment of breast cancer from the direction of molecular regulatory network, providing reference for the development of new drugs and the compatibility of effective components.
