摘要
目的探讨白介素-6(interleukin-6,IL-6)、白介素-17(interleukin-17,IL-17)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)与高血压模型大鼠肾纤维化的相关性。方法将原发性高血压模型大鼠20只纳入观察组,同时将野生型健康大鼠20只纳入对照组,比较两组大鼠血浆中IL-6、IL-17、TGF-β1水平以及两组大鼠肾脏中IL-6、IL-17、TGF-β1mRNA的表达情况。采用Masson染色法比较两组大鼠肾纤维化程度,并分析原发性高血压模型大鼠肾纤维化程度与IL-6、IL-17、TGF-β1的相关性。结果观察组大鼠血浆中IL-6和IL-17水平、肾脏中IL-6和IL-17表达量及肾脏中IL-6和IL-17 mRNA相对表达量均显著高于对照组(均P<0.01);血浆中TGF-β1水平、肾脏中TGF-β1表达量及肾脏中TGF-β1 mRNA相对表达量均显著低于对照组(均P<0.05)。高血压模型大鼠肾纤维化程度与血浆中IL-6和IL-17水平、肾脏中IL-6和IL-17表达量及肾脏中IL-6和IL-17 mRNA相对表达量均呈正相关(均P<0.05);与肾脏中TGF-β1、TGF-β1 mRNA相对表达量均呈负相关(均P<0.05)。结论在高血压模型大鼠血浆和肾脏中,IL-6和IL-17的表达均升高,TGF-β1的表达均降低,且均与肾纤维化有关。
Objective To investigate the relationship between interleukin-6(IL-6), interleukin-17(IL-17), transforming growth factor-β1(TGF-β1) and renal fibrosis in hypertensive rats. Method 20 essential hypertension model rats were included in observation group, and 20 wild type healthy rats were included in control group. The levels of IL-6, IL-17 and TGF-β1 in plasma and the expression of IL-6, IL-17 and TGF-β1 m RNA in kidney were compared between the two groups. The degree of renal fibrosis in the two groups was compared by Masson staining, and the correlation between the degree of renal fibrosis and IL-6, IL-17, TGF-β1 in essential hypertension model rats was analyzed. Result The levels of IL-6 and IL-17 in plasma, the expression of IL-6 and IL-17 in kidney and the relative expression of IL-6 and IL-17 m RNA in kidney in observation group were significantly higher than those in control group(all P<0.01), the level of TGF-β1 in plasma, the expression of TGF-β1 in kidney and the relative expression of TGF-β1 m RNA in kidney were significantly lower than those in control group(all P<0.05). The degree of renal fibrosis was positively correlated with the levels of IL-6 and IL-17 in plasma, the expression of IL-6 and IL-17 in kidney and the relative expression of IL-6 and IL-17 m RNA in kidney(all P<0.05), and negatively correlated with the relative expression of TGF-β1 and TGF-β1 m RNA in kidney(all P<0.05). Conclusion In plasma and kidney of hypertensive rats, the expression of IL-6 and IL-17 is increased, the expression of TGF-β1 is decreased, which is related to renal fibrosis.
