珠子参总皂苷对大鼠成骨细胞的保护作用及其机制初探 被引量:3

摘要 离体培养大鼠原代成骨细胞,分别采用MTT法、EdU成像法、测定碱性磷酸酶法、茜素红染色法考察了不同浓度珠子参总皂苷对成骨细胞活力、增殖、分化、矿化等的影响;并采用Western bloting法研究了不同浓度珠子参总皂苷对成骨细胞OPG和RANKL蛋白表达的影响。结果表明,珠子参总皂苷浓度为100μg·mL^-1时可提高成骨细胞活力;浓度为50μg·mL^-1、100μg·mL^-1、200μg·mL^-1时可显著促进成骨细胞增殖、分化和矿化;珠子参总皂苷呈浓度依赖性地提高OPG/RANKL相对表达比值。 We cultured rat primary osteoblasts in vitro,and investigated the effects of total saponins from Panax japonicas on activity,proliferation,differentiation,and mineralization of osteoblasts by MTT,EdU image construction,alkaline phosphatase assay,and Alizarin Red staining,respectively.Moreover,we studied the effects of total saponins from Panax japonicas on the protein expression of OPG and RANKL in osteoblasts by Western blotting.The results show that the activity of osteoblasts significantly increases when the concentration of total saponins is 100 μg·mL^-1.Meanwhile,the proliferation,differentiation,and mineralization of osteoblasts significantly increase when the concentrations of total saponins are 50 μg·mL^-1,100 μg·mL^-1,and 200 μg·mL^-1.Furthermore,the expression ratio of OPG/RANKL significantly increases in a dose-dependent manner within total saponins of Panax japonicas.
