miR-31在COPD患者外周血中的表达及其临床意义

目的 检测慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMCs)中微小RNA-31(miR-31)表达量并探讨其临床意义。方法 采用实时荧光定量聚合酶链反应(qPCR)的方法检测38例COPD患者(COPD组)、15例急性支气管炎(AB)患者(AB组)和40例体检健康者(对照组)PBMCs中miR-31的表达,并对15例治疗后患者进行跟踪检测。酶联免疫吸附测定法(ELISA)检测COPD患者血浆中白细胞介素4(IL-4)和γ-干扰素(IFN-γ)水平。结果 COPD 患者PBMC中miR-31的表达明显高于对照组和AB组,AB组PBMC中miR-31的表达明显高于对照组;COPD患者治疗后好转后miR-31的表达明显下降;COPD患者临床症状严重者其PBMC中miR-31的表达更高,COPD患者PBMC中miR-31的表达与血浆CRP水平呈负相关( r =-0.663 8, P <0.05),COPD患者PBMC中miR-31的表达与血浆中IL-4水平呈负相关( r =-0.476 4, P < 0.05 )。结论 COPD患者PBMC中miR-31的表达参与COPD的发生与发展,其可能在Th2类细胞中起重要作用。

Objective To investigate the expression of miR-31 in peripheral blood mononuclear cells(PBMCs) of patients with chronic obstructive pulmonary disease(COPD) and explore its clinical significance. Methods The expression of miR-31 in PBMCs from 38 patients with COPD(COPD group),15 patients with acute bronchitis(AB group),40 Healthy persons (control group) and 15 patients treated after tracking were measured by qPCR.The plasma level of IL-4 and IFN-γ in patients with COPD were measured by enzyme linked immunosorbent assay. Results The expression of miR-31 in PBMC of patients with COPD was higher than AB group and control group,the expression of miR-31 in PBMC of patients with AB group were higher than control group,and it was decreased after treatment in patients with COPD;The expression of miR-31 in serious patients with COPD was higher than the other patients,the expression of miR-31 in patients with COPD was negative correlated with CRP level( r =-0.663 8, P <0.05). The expression of miR-31 in patients with COPD was negative correlated with the level of IL-4 in plasma( r =-0.476 4, P <0.05). Conclusion The expression of miR-31 in patients with COPD is significantly increased,which is related with the occurrence and development of COPD,and it is probably to impact the function of Th2 type cells.
