代谢综合征合并阻塞性睡眠呼吸暂停低通气综合征患者CDKN2A/2B基因多态性的研究 被引量:2

STUDY ON CDKN2A/2B GENE POLYMORPHISM IN PATIENTS WITH METABOLIC SYNDROME AND OBSTRUCTIVE SLEEP APNEA-HYPOPNEA SYNDROME

摘要 目的探讨细胞周期蛋白依赖激酶抑制剂(cyclin-dependent kinase inhibitor 2A/2B,CDKN2A/2B)基因多态性与代谢综合征合并阻塞性睡眠呼吸暂停低通气综合征患者的关系。方法选取2017年6月—2018年11月我院门诊就诊的216例代谢综合征患者,根据多导睡眠仪诊断为阻塞性睡眠呼吸暂停低通气综合征患者116例为观察组,单纯代谢综合征患者100例为代谢综合征对照组,选取同期我院健康查体者98例为健康对照组。分别采用聚合酶链反应限制性片段长度多态性测定三组患者CDKN2A/2B基因rs10811661基因型分布及频率。结果三组患者CDKN2A/2B基因rs10811661的TT、TC、CC三种基因型频率分布比较:与健康对照组比较,代谢综合征对照组和观察组三种基因型频率分布差异均有统计学意义(P<0.05),与代谢综合征对照组比较,观察组三种基因型频率分布差异无统计学意义(P>0.05)。三组患者CDKN2A/2B基因rs10811661的T、C两种等位基因频率分布比较:与健康对照组比较,代谢综合征对照组和观察组T、C两种等位基因频率分布差异均有统计学意义(P<0.05),其中观察组等位基因T分布频率明显高于健康对照组(P<0.05),与代谢综合征对照组比较,观察组等位基因型频率分布差异无统计学意义(P>0.05)。结论CDKN2A/2B基因rs10811661多态性可能与代谢综合征合并阻塞性睡眠呼吸暂停低通气综合征的发病有关,存在一定的易感性。 Objective To investigate the relationship between CDKN2 A/2 B gene polymorphism and patients with metabolic syndrome and obstructive sleep apnea hypopnea syndrome.Methods From January 2016 to December 2017,two hundred and sixteen patients with metabolic syndrome in Affiliated Hospital of North China University of Science and Technology were selected.116 patients who were diagnosed with obstructive sleep apnea-hypopnea syndrome according to polysomnography were selected as observation group,100 patients with pure metabolic syndrome were selected as control group of metabolic syndrome,and 98 cases of health examination during the same period in our hospital were selected as healthy control group.The distribution and frequency of CDKN2 A/2 B gene rs10811661 genotype were determined by polymerase chain reaction restriction fragment length polymorphism.Results The distribution of TT,TC and CC genotypes of CDKN2 A/2 B gene rs10811661 was significantly different between control group and control group of metabolic syndrome(P<0.05).The frequency of T allele distribution in the control group of metabolic syndrome was significantly higher than that in the control group(P<0.05).The distribution of the three genotypes in the observation group was significantly higher than that in the control group(P<0.05).The frequency of T allele distribution in observation group was significantly higher than that of control group(P<0.05).There was no significant difference in genotype and allele frequency distribution between the two groups(P>0.05).Conclusion The rs10811661 polymorphism of CDKN2 A/2 B gene may be associated with the development ofmetabolic syndrome with obstructive sleep apnea hypopnea syndrome and have a certain susceptibility.
