半枝莲对5-FU治疗晚期大肠癌的疗效及血清miRNA-34a表达水平的影响

Effects of Sculellaria Barbata on 5-FU-Treated Advanced Colorectal Cancer and Serum miRNA-34a Levels

目的 探讨半枝莲对5-氟尿嘧啶(5-FU)治疗晚期大肠癌的临床疗效及血清miRNA-34a表达水平的影响。方法 将60例晚期大肠癌患者按随机数字表法分为对照组和半枝莲组,每组30例。对照组给予5-FU治疗,半枝莲组在对照组治疗的基础上给予半枝莲片口服。比较2组血清miRNA-34a表达水平及治疗有效率、患者的生存期,采用Pearson相关系数分析血清miRNA-34a表达水平与预后的关系。结果 半枝莲组治疗有效率明显高于对照组(66.67%比40.00%,P <0.05),半枝莲组患者的生存期为(21.27±2.54)个月,明显长于对照组的(18.82± 3.44)个月(P<0.05),半枝莲组血清miRNA-34a表达水平为(1.21±0.33),明显高于对照组的(1.03±0.25)(P<0.05)。相关性分析结果显示,血清miRNA-34a表达水平与患者的生存期呈正相关(r=0.78,P <0.05)。结论 半枝莲能提高5-FU治疗晚期大肠癌的治疗有效率,延长患者的生存期,这可能与半枝莲下调血清miRNA-34a的表达水平有关。

Objective To explore the effects of Sculellaria barbata on clinical efficacy and serum miRNA-34a levels in the treatment of advanced colorectal cancer with 5-fluorouracil(5-FU).Methods Sixty patients with advanced colorectal cancer were randomly treated with 5-FU alone(control group,n=30) or in combination with oral Sculellaria barbata tablets(Sculellaria barbata,n=30).Serum miRNA-34a levels,therapeutic efficiency and survival period were compared between the two groups.Pearson correlation coefficient was used to analyze the relationship between serum miRNA-34a expression and prognosis.Results Compared with control group,Sculellaria barbata treatment increased therapeutic efficiency(66.67% vs 40.00%,P<0.05),prolonged survival period((21.27±2.54) months vs(18.82± 3.44) months,P<0.05),and elevated serum microRNA-34a levels((1.21±0.33) vs(1.03+0.25),P<0.05).The correlation analysis showed that serum microRNA-34a expression was positively correlated with survival period(r=0.78,P<0.05).Conclusion Sculellaria barbata can improve therapeutic efficiency and prolong survival period in the treatment of advanced colorectal cancer with 5-FU.The mechanism of Sculellaria barbata action may be involved in the down-regulation of serum microRNA-34a expression.
