摘要 目的探讨大蒜提取物S-烯丙基-L-半胱氨酸对辐射损伤小鼠造血系统和抗氧化功能的防护作用。方法利用造血系统和抗氧化功能实验,小鼠全身照射至吸收剂量为7.5 Gy后测外周血白细胞计数(WBC)、股骨有核细胞数(BMNC)、骨髓DNA含量,观察小鼠肝脏中含量的变化。结果与单纯照射组相比,照射给药中剂量组的WBC、BMNC、骨髓DNA含量均显著升高,分别从(0.17±0.05)×10^9/L,(0.56±0.07)×10^6/根,0.21±0.14升高至(0.67±0.08)×10^9/L,(4.45±1.76)×10^6/根,0.58±0.12(P<0.05);通过对肝脏中MDA,GSH,CAT,T-SOD的含量测定,发现照射前给予SAC能一定程度上抑制辐射引起的MDA升高及GSH,CAT,T-SOD降低的幅度(P<0.05)。结论大蒜提取物S-烯丙基-L-半胱氨酸对辐射损伤小鼠的抗氧化功能和造血系统具有较强的防护作用。 Objective To investigate the protective effect of the garlic extract such as S-allyl-L-cysteine on the hematopoietic system and anti-oxidative function in radiation-injured mice.Methods Hematopoietic system and anti-oxidative function experiments of mice were carried out to measure the peripheral white blood cell(WBC),bone marrow nucleated cells(BMNC)and bone marrow DNA content of mice after the whole body was irradiated to the absorbed dose of 7.5 Gyγ-rays.The changes in the above indexes in the liver of mice were observed.Results Compared with the irradiation group,the WBC,BMNC and bone marrow DNA content of mice administered with middle dose group increased from(0.17±0.05)×10^9/L,(0.56±0.07)×10^6 per femur,0.21±0.14 to(0.67±0.08)×10^9/L,(4.45±1.76)×10^6 per femur and 0.58±0.12(P<0.05).By measuring the contents of malondialdehyde(MDA),reduced glutathione(GSH),catalase(CAT)and total superoxide dismutase(T-SOD)in the liver,it was found that the administration of SAC before irradiation can inhibit the increase of MDA and the decrease of GSH,CAT and T-SOD caused by radiation to some extent(P<0.05).Conclusion S-Allyl-L-cysteine has protective effect on the hematopoietic system and anti-oxidative function in radiation-injured mice.
