保健品中原花青素含量检测方法的比较研究 被引量:6

摘要 为了探讨不同分析方法对保健品中原花青素含量检测的精准度,对现有标准中原花青素测定的前处理方法进行优化,并分别选用高效液相色谱法和铁盐催化比色法对保健品中原花青素的测定进行方法学对比和验证。前处理优化为用顶空瓶于烘箱100℃加热40min,分析方法选用高效液相色谱法优于铁盐催化比色法。结果显示,高效液相色谱法可在6min内完成对目标物的分离,线性方程为Y=540.7X+3694.23,r^2=0.9997,方法回收率为99.0%~103.6%,相对标准偏差(RSD)均小于2.2%,方法检出限为1.5mg/kg,方法定量限为5.0mg/kg,测定结果比铁盐催化比色法更精准、稳定。由此得出,高效液相色谱法测定保健品中原花青素的含量回收率及灵敏度更高,准确度和稳定性更好,更具有推广意义。 In order to explore the accuracy of different analytical methods for the determination of proanthocyanidins in health care products,the pretreatment methods for the determination of proanthocyanidins in existing standards are optimized,and the methodological comparison and validation of the determination of proanthocyanidins in health care products by high performance liquid chromatography and iron salt catalytic colorimetry are carried out respectively.The pretreatment is optimized by heating the oven with headspace bottle at 100℃for 40 minutes.The analytical method of high performance liquid chromatography is superior than that of iron salt catalytic colorimetry.The results show that the separation of the target by high performance liquid chromatography could be completed within 6 minutes.The linear equation is Y=540.7X+3 694.23,r^2=0.999 7.The recovery of the method is 99.0%-103.6%.The relative standard deviation(RSD)is less than 2.2%.The detection limit of the method is 1.5 mg/kg.The quantitative limit of the method is 5.0 mg/kg.The determination result is more accurate than that of iron salt catalytic colorimetry.It is concluded that the determination of proanthocyanidins in health products by high performance liquid chromatography has higher recovery and sensitivity,better accuracy and stability,and has more positive significance for promotion.
