摘要
壳聚糖(chitosan,COS)结构中的—NH 2和—OH具有良好的金属离子配位能力,有效地络合钙离子后有望成为具备无机/有机双重特性的生物材料。采用直接吸附法合成壳聚糖钙(chitosan-Ca,COS-Ca)及羧甲基壳聚糖钙(carboxymethyl chitosan-Ca,CM-COS-Ca)配合物,在对产物光谱学性能研究的基础上,探讨壳聚糖及其衍生物钙配合物对成骨细胞功能及细胞因子分泌的影响。结果表明,钙离子与壳聚糖和羧甲基壳聚糖(carboxymethyl chitosan,CM-COS)发生络合,所形成的钙配合物可促进成骨细胞的增殖和分化,增强成骨细胞OPG蛋白的表达,同时抑制RANKL蛋白的表达。本研究对后续材料功能的研究和临床应用均具有指导意义。
The-NH2 and-OH groups in chitosan(COS)have good metal ion coordination ability.It is expected that the chitosan can be used as biological materials with inorganic/organic properties after complexing Ca^2+effectively.In this study,chitosan-Ca(COS-Ca)complexes and carboxymethyl chitosan-Ca(CM-COS-Ca)complexes were synthesized by direct adsorption and focused on the spectral properties.Furthermore,the effects of COS-Ca and CM-COS-Ca on the function and cytokine secretion of osteoblast MC3T3-E1 were investigated.The results show that calcium ion can be complexed with chitosan and carboxymethyl chitosan.COS-Ca and CM-COS-Ca have good cell compatibility,promoting the proliferation and differentiation of osteoblasts.COS-Ca and CM-COS-Ca can enhance the expression of OPG protein and inhibit the expression of RANKL protein in osteoblasts,promoting osteogenesis.This study has guiding significance for the study of material function and clinical application.
