血清circRNA与甲状腺乳头状癌的相关性研究

摘要 目的探讨血清circRNA与甲状腺乳头状癌的相关性。方法选择2018年11-12月在新疆医科大学第一附属医院甲状腺外科行甲状腺切除术,并经病理证实的结节性甲状腺肿(nodular goiter, NG)患者(NG组)和PTMC患者(PTMC组)各13例,均为女性。收集研究对象的一般资料。术前收集患者晨起空腹上肢静脉血5 mL,离心得血清。2组各选取3名患者进行circRNA芯片检测,后再各选取3例对circRNA芯片检测结果进行验证,2组各剩余7例,对血清样本进行circRNA扩大样本验证。结果经芯片检测NG与PTMC患者中血清筛选差异表达circRNA,显示差异表达上调的有14个,下调的有6个。经Go软件检测2组患者血清中的差异表达circRNA的亲本基因分析,得出富集前10位的生物学过程、细胞组分和分子功能。NG患者与PTMC患者血清中的差异表达circRNA相关的基因参与了Olfactory transduction通路(37个基因)。RT-PCR验证表明hsacircRNA001359,hsacircRNA003416,hsacircRNA104982和hsacircRNA043614可能可以作为PTMC诊断的生物学标记物,其中hsacircRNA001359表达水平显著上调5.8倍(P<0.01)。结论血清circRNA001359可能可以做为诊断女性、甲状腺结节患者术前诊断PTMC性质的生物学标记物,后期需要进一步扩大样本量验证。 Objective To investigate the correlation between serum circRNA and thyroid papillary carcinoma. Methods From November to December 2018, 13 patients with nodular goiter(NG) and 13 patients with PTMC(PTMC) were selected for thyroidectomy in the Thyroid Surgery Department of the First Affiliated Hospital of Xinjiang Medical University. The general data of the subjects were collected. Before operation, 5 mL of fasting venous blood of upper limb was collected and serum was obtained by centrifugation. Three patients in each group were selected for circRNA chip detection, and then three patients in each group were selected to verify the results of circRNA chip detection. The remaining 7 patients in each group were selected to verify the enlarged circRNA sample in serum samples. Results The differential expression of circRNA was detected by microarray in serum of patients with NG and PTMC. The results showed that the differential expression of circRNA was up-regulated in 14 cases and down-regulated in 6 cases. The genetic analysis of parents with differential expression of circRNA in serum of the two groups was performed by Go software, and the biological process, cell components and molecular functions of the top 10 enrichment sites were obtained. The differential expression of circRNA-related genes in the serum of NG patients and PTMC patients are involved in the Olfactory transduction pathway(37 genes). RT-PCR confirmed that hsacircRNA001359, hsacircRNA003416, hsacircRNA104982 and hsacircRNA043614 might be used as biomarkers for PTMC diagnosis, and the expression level of hsacircRNA001359 was significantly increased by 5.8 times(P<0.01). Conclusion Serum circRNA001359 may be used as a biological marker for preoperative diagnosis of PTMC in women and patients with thyroid nodules, and further sample size verification is needed in the future.
