摘要 目的探究神经菌毛素-1(Neuropilin-1,NRP-1)对舌癌TCa8113细胞增殖、迁移能力、细胞凋亡及对化疗药物敏感性的影响。方法以舌癌TCa8113细胞为研究对象,分别使用shRNA方法构建NRP-1敲低TCa8113细胞株和使用NRP-1单克隆抗体两种方法抑制NRP-1功能,通过对比抑制NRP-1功能后TCa8113细胞株与正常TCa8113细胞株,确定NRP-1对TCa8113细胞的增殖、迁移能力、细胞凋亡及对化疗药物敏感性的影响。结果成功构建NRP-1敲低TCa8113细胞株(NRP-1-knockdown)。NRP-1低表达细胞株和NRP-1单克隆处理抑制NRP-1功能后,细胞的增殖、迁移能力与正常TCa8113细胞株相比均显著降低(P<0.05),而细胞凋亡及对化疗药物的敏感性增加(P<0.05)。结论NRP-1可促进舌癌TCa8113细胞增殖和细胞迁移能力,抑制细胞的凋亡,降低细胞的化疗敏感性。 Objective To investigate the effects of neuropilin-1(NRP-1)on proliferation,migration,apoptosis of tongue cancer cell line TCa8113 and the sensitivity to chemotherapeutic drugs.Methods The tongue cancer TCa8113 cells were used as the research objects.NRP-1 knockdown TCa8113 cell line by shRNA and NRP-1 monoclonal antibody were used to inhibit NRP-1 function.After TCa8113 cells were inhibited by NRP-1 function,the proliferation,migration,apoptosis and sensitivity of TCa8113 cells to chemotherapeutic drugs were observed and compared with the normal TCa8113 cell line.Results NRP-1 knockdown TCa8113 cell line(NRP-1 knockdown)was successfully constructed.In NRP-1 low-expression cell line or after NRP-1 monoclonal antibody treatment,the proliferation and migration ability of the cells were significantly decreased compared with the normal TCa8113 cell line(P<0.05),while the apoptosis and sensitivity to chemotherapy drugs were increased(P<0.05).Conclusion NRP-1 can promote the proliferation and cell migration of tongue cancer TCa8113 cells,inhibit cell apoptosis and reduce the chemosensitivity of cells.
