摘要
目的探讨过敏性紫癜患儿外周血miR-155表达及与Th17/Treg平衡的关系。方法选取79例过敏性紫癜患儿为研究对象,60例健康体检儿童作为对照,采用qRT-PCR技术检测外周血miR-155、RORγt和Foxp3mRNA表达,流式细胞术检测Th17和Treg细胞比例,酶联免疫吸附法检测细胞因子IL-6、IL-17、IL-22、IL-23、IL-10和TGF-β1表达,Pearson相关性分析miR-155与Th17和Treg细胞,RORγt和Foxp3mRNA,细胞因子IL-6、IL-17、IL-22、IL-23、IL-10和TGF-β1表达之间关系。结果与健康对照相比,过敏性紫癜患儿外周血miR-155、Th17细胞比例、转录因子RORγt、相关细胞因子IL-6、IL-17、IL-22和IL-23均显著升高(P<0.05),Treg细胞比例、转录因子Foxp3、相关细胞因子IL-10和TGF-β1均显著降低(P<0.05)。miR-155与Th17细胞比例、RORγt、IL-6、IL-17、IL-22和IL-23均呈正相关(P<0.05),与Treg细胞比例、Foxp3、IL-10和TGF-β1均呈负相关(P<0.05)。结论过敏性紫癜患儿外周血miR-155高表达,Th17/Treg平衡失调,miR-155可能通过调节Th17/Treg平衡参与过敏性紫癜的疾病过程,是该疾病潜在的治疗靶点。
Objective To investigate the expression of miR-155 in peripheral blood of children with Henoch-Schonlein purpura and its relationship with Th17/Treg balance.Methods A total of 79 children with Henoch-Schonlein purpura were selected as study group and 60 healthy children as control group.The expression of miR-155,RORγT and Foxp3 in peripheral blood were detected by qRT-PCR,and the proportions of Th17 and Treg cells were detected by flow cytometry,and the expression of cytokines IL-6,IL-17,IL-22,IL-23,IL-10,and TGF-β1 was detected by ELISA.Pearson correlation analysis was used to analyze the relationships between the expression of miR-155 and Th17 and Treg cells,RORγT and Foxp3 mRNA,cytokines IL-6,IL-17,IL-22,IL-23,IL-10 and TGF-β1.Results Compared with healthy controls,the percentage of Th17 cells,and the levels of miR-155,transcription factor RORγt,related cytokines IL-6,IL-17,IL-22,and IL-23 in peripheral blood of children with Henoch-Schonlein purpura increased significantly(P<0.05),while Treg cell proportion,and the levels of transcription factor Foxp3,related cytokines IL-10,and TGF-β1 were significantly decreased(P<0.05).The miR-155 was positively correlated with Th17 cell ratio,RORγt,IL-6,IL-17,IL-22 and IL-23(P<0.05),but negatively correlated with Treg cell proportion,Foxp3,IL-10 and TGF-β1(P<0.05).Conclusion The expression of miR-155 in peripheral blood of children with Henoch-Schonlein purpura is high with the imbalance of Th17/Treg.The miR-155 may be involved in the disease process of Henoch-Schonlein purpura by regulating the Th17/Treg balance,which is a potential therapeutic target for the disease.
