摘要
在盐稻8号辐射诱变后代中筛选出1个黄叶突变体,并命名为yl1。遗传分析表明,黄叶表型为单隐性核基因控制。通过对yl1的表型观察、光合色素含量测定、叶绿体超微结构观察和叶绿素生物合成途径相关基因的表达分析发现,突变体yl1全生育期均表现黄叶,叶绿素含量显著降低,叶绿体缺少内囊体片层结构,并且大部分叶绿素生物合成途径的基因表达显著降低。本研究结果可为下一步突变基因定位和基因功能研究打下良好基础。
In this study,we selected a yellow leaf mutant of rice named yl1 in the late generation of the rice cultivar 'Yandao 8';it was generated by induction with 60Co-γ ray radiation.The genetic analysis showed that the yellow leaf phenotype of yl1 was controlled by a single recessive nuclear gene.We observed its phenotype,determined the contents of photosynthetic pigments in its leaves,observed the ultrastructure of its chloroplast,and analyzed the expression patterns of the genes involved in the chlorophyll biosynthetic pathway in its leaves.The leaves of the mutant remained yellow during the whole growth period,its leaf chlorophyll contents decreased significantly compared with those of the wild type,its chloroplast lacked endocyst lamellar structure,and most of the genes involved in its chlorophyll biosynthetic pathway were expressed at a significantly lower level than the counterparts of the wild type.The results lay a solid foundation for mapping the mutated gene and deciphering its function in the future study.
