摘要
为博落回提取血根碱开发新原料,为工业化生产提供依据。在单因素试验的基础上,以血根碱转移率为评价指标,分别对博落回茎叶使用酸性乙醇回流和酸水渗漉法进行工艺研究。试验结果表明酸性乙醇回流提取工艺最佳条件为乙醇体积分数90%,液固比30 1 ml/g,硫酸浓度0.12%,提取温度80 ℃,提取时间2 h,血根碱转移率达到(89.36±0.49)%(n=5, R2=0.54%),最终得到博落回茎叶提取物收率(24.27±0.92)%(n=5,R2=3.77%),血根碱含量(1.65±0.06)%(n=5,R2=3.36%);酸水渗漉提取工艺最佳条件为硫酸浓度1%,浸泡时间30 min,渗漉温度80 ℃,溶剂用量30倍,渗漉速度0.5 ml/min,血根碱转移率达到(83.63±2.09)%(n=5, R2=2.49%),最终得到博落回茎叶提取物收率(21.64±0.78)%(n=5,R2=3.61%),血根碱含量(2.25±0.11)%(n=5, R2=4.88%)。两种提取方法中酸水渗漉法碱沉步骤简便,得到的提取物中血根碱的含量比酸性乙醇回流法高出36.36%,溶剂用量一致,在工业化生产中应结合实际情况进一步优化参数条件,为博落回茎叶开发利用提供了理论依据。
For developing new materials and of. fer a sound evidence for the mass production in. dustry, the extraction process of sanguinarine from the stem and leaf of Macleaya cordata was studied in this work. On the basis of single fac. tor experiment, the factors affecting the transfer rate of sanguinarine investigated by ethanol reflux extraction and percolation extraction. The experiment results indicated that the optimum conditions for the ethanol reflux extraction were obtained as follows:the ethanol concentration was 90%, liquid-solid ratio was 30:1, sulfuric acid concentration was 0.12%, extraction temperature was 80 ℃ and extraction time was 2 h. Under the conditions, the transfer rate of sanguinarine was (89.36±0.49)%(n=5, R2=0.54%), the content of sanguinarine in the extract obtained from the final is (1.65±0.06)%(n= 5, R2=3.36%), the yield was (24.27±0.92)%(n=5, R2=3.77%), and the percolation extraction as follows, sulfuric acid concentration was 1%, soaking time was 30 min, percolation temperature was 80 ℃, liquid-solid ratio was 30:1ml/g and percolation rate was 0.5 ml/min. Under the conditions, the transfer rate of sanguinarine was (83.63±2.09)%(n=5, R2=2.49%), the content of sanguinarine in the final extract was (2.25±0.11)%(n=5, R2=4.88%), the yield was (21.64±0.78)%(n=5, R2=3.61%). In the two extraction methods, the acid water percolation method has a simple alkali precipitation step than acid ethanol reflux extraction. The content of sanguinarine in the extracts was 36.36% higher than acid ethanol reflux method, and the solvent dosage is consistent. In the industrial production, the parameters should be controlled according to the actual situation. The research can provide data for the stem and leaf of Macleaya cordata's development and utilization.
