摘要
目的观察高密度脂蛋白模拟肽(Rev-D4F)对肿瘤坏死因子(TNF)-α诱导的内皮祖细胞凋亡和黏附的影响。方法差速贴壁法培养内皮祖细胞;吸附低度脂蛋白和结合凝集素方法鉴定内皮祖细胞;分别给予Rev-D4F(50μg/ml)和TNF-α(30ng/ml)进行处理,采用黏附测定实验和流式检测内皮祖细胞的黏附功能和凋亡情况。结果TNF-α促进内皮祖细胞凋亡,但TNF-α对内皮祖细胞黏附并没有作用,Rev-D4F预先干预能明显抑制TNF-α诱导的内皮祖细胞凋亡,提高内皮祖细胞黏附功能。结论Rev-D4F抑制TNF-α诱导的内皮祖细胞凋亡,提高内皮祖细胞黏附功能。
Objective To study the effects of high density lipoprotein mimic peptide Rev-D4F on the apoptosis and adhesion of endothelial progenitor cells(EPCs)induced by tumor necrosis factor(TNF)-α.Methods EPCs were cultured by differential time attachment and identified by adsorption of low lipoprotein and binding lectin.EPCs were treated with Rev-D4F(50μg/ml)and TNF-α(30 ng/ml)respectively,the adhesion function and apoptosis of EPCs were measured by adhesion assay and flow cytometry.Results TNF-αpromoted EPCs apoptosis,but had no effect on EPCs adhesion.Rev-D4F pretreatment significantly inhibited TNF-αinduced EPCs apoptosis and improved endothelial progenitor cell adhesion function.Conclusions Rev-D4F could inhibit the apoptosis of EPCs induced by TNF-αand improve the adhesion function of EPCs.
