摘要
本研究利用Real-timePCR分析和比较sox9与amh基因在雄、雌虹鳟Oncorhynchus mykiss及及其伪雄鱼各组织中的表达模式。结果表明:sox9基因在这三种鱼性腺、脑、脾、肝以及脑垂体中的表达显著高于其他组织(P<0.05)。在性腺组织中,sox9基因在雄鱼中的表达量显著高于伪雄鱼(P<0.01),而在伪雄鱼中的表达量又显著高于普通雌鱼(P<0.01)。脑组织中,雌鱼sox9的表达显著高于雄鱼和伪雄鱼(P<0.05)。Amh基因在性腺和脑组织中的表达显著高于其他组织(P<0.05)。该基因在伪雄鱼性腺和脑中的表达均显著高于雄鱼和雌鱼(P<0.05)。结果表明,外源雄激素的诱导可导致伪雄虹鳟性腺和脑组织中sox9和amh基因的表达显著高于雌性虹鳟,促进其性腺的雄性化发育。本研究可为全雌性虹鳟制种技术研究奠定重要理论基础。
Expression profiles of sox9 and amh genes detected in different tissues of male, female and pseudo-male rainbow trout (Oncorhynchus mykiss) by Real-time PCR. Results showed that expressions of sox9 were significantly higher in gonad, brain, spleen, liver and pituitary than that in other tissues (P < 0.05 or P < 0.01). In the tissue of gonads, sox9 expression in normal males was significantly high than pseudo-males (P < 0.01), very significantly higher than that in normal females (P < 0.01). In brains, expression of sox9 in female was significantly higher than that in male and pseudo-male (P < 0.05).There were very significantly or significantly higher Amh expression levels in gonads and brains than that in other tissues (P < 0.01 or P < 0.05). In addition, expression of amh has be found to be significantly higher in gonad and brain of pseudo-male than that in normal males and females. We can draw a conclusion that the expression of sox9 and amh were elevated significantly by exogenous hormone induction, as the same case in male-like development. The findings establish an important theoretic ground for the study of all-female production techniques.
