摘要
Viroporins是一类由病毒编码的小分子疏水性跨膜蛋白,具有1个~3个跨膜结构域,通过寡聚化形成离子通道,主要通过其离子通道活性来增加膜的渗透性,进而影响细胞运输、离子通量以及细胞器之间的连接和交换等基本生物学过程。病毒编码的Viroporins不仅与病毒致病性有关,还参与调节宿主细胞的电化学平衡,并影响病毒的复制,因此它们有望成为抗病毒治疗的靶点。不同病毒的Viroporins在结构上具有不同的特点以及对病毒复制的影响、诱导的宿主细胞生物学反应,包括细胞自噬、细胞凋亡、细胞焦亡等方面具有不同的作用。随着生物技术的发展,Viroporins的结构和功能被研究的越来越透彻,为发现新的和更有效的病毒感染抑制剂提供参考。
Viroporins are small transmembrane viral proteins with ion channel activities modulating properties of intracellular membranes,which impacts several fundamental biological processes such as trafficking,ion fluxes as well as connections and exchanges between organelles.Viral Viroporins play an important role either in regulating virus replication,pathogenicity or modulating the electrochemical balance in the subcellular compartments of host cells,so they are expected to become targets for antiviral therapy.Viroporins of different viruses have different functional properties in structure,as well as effects on viral replication and induced host cell biological responses,including autophagy, apoptosis, and pyroptosis.With the development of biotechnology,the structure and function of Viroporins have been studied more and more thoroughly,providing a theoretical basis for discovering new and more effective viral infection inhibitors and new therapeutic methods for studying pathogens.
