摘要
目的观察电针对造影剂肾病模型(CIN模型)大鼠肾损伤的影响。方法选取雄性清洁级SD大鼠50只,随机选取10只大鼠为对照组,将剩余40只制备成糖尿病模型,造模成功30只,随机分为模型组,电针组,冬虫夏草组,各10只,糖尿病模型建好后饲养9周,对照组与模型组给予等量0.9%氯化钠注射液灌胃,电针组给予电针治疗,冬虫夏草组给予冬虫夏草菌粉[3 g/(kg·d)]灌胃,各组干预1周后建立糖尿病CIN模型,处死大鼠,摘取左肾,以备肾脏病理研究,摘取右肾,用于Western blotting定量分析,观察各组大鼠的尿素氮(BUN)、血肌酐(SCr)、氧化应激指标及肾组织Caspase-3、Caspase-9蛋白检测水平,并采用HE染色和TUNEL染色观察各组肾脏组织的病理改变。结果1)与模型组比较,冬虫夏草组、电针组的大鼠的BUN、Scr、Kim-l、NGAL均明显下降(P<0.05)。2)与模型组比较,冬虫夏草组、电针组氧化应激指标均降低,以及Caspase-9、Caspase-3表达水平均升高(P<0.05)。3)其中冬虫夏草组、电针组差异无统计学意义(P>0.05)。结论电针能够减轻糖尿病大鼠CIN的早期肾损害,且与冬虫夏草治疗效果相当。
Objective:To observe the effect of electro-acupuncture on renal injury in rats with contrast-induced nephropathy(CIN).Methods:50 male clean SD rats were selected and 10 rats were randomly selected as the blank control group.The remaining 40 rats were made into diabetic model.30 rats were successfully modeled and randomly divided into the model group,electro-acupuncture group,Cordyceps sinensis group,10 in each.After 9-week feeding,blank control group and model group were given the same amount of saline,group A electroacupuncture treatment,Cordyceps sinensis group cordyceps sinensis fungus powder[3 g/(kg·d)].After one week of intervention,the CIN model of diabetes mellitus was established.Rats were executed and left kidney was taken for pathological study,right kidney for quantitative analysis by Western blotting.BUN,Scr,oxidative stress index,caspase 3 and caspase 9 protein in kidney tissues were observed,and the pathological changes of kidney tissues were observed by HE staining and TUNEL staining.Results:Compared with the model group,BUN,Scr,Kim-l and NGAL in Cordyceps sinensis group and group A decreased significantly(P<0.05).Compared with model group,the oxidative stress index in Cordyceps sinensis group and electro-acupuncture group decreased,and the expression levels of caspase 9 and caspase 3 were higher;the difference was statistically significant(P<0.05).There was no significant difference between electro-acupuncture group and Cordyceps sinensis group(P>0.05).Conclusion:Electro-acupuncture can alleviate the early renal damage of CIN in diabetic rats,and the therapeutic effect is similar to that of Cordyceps sinensis.
