二甲双胍对黑色素瘤A375细胞生长和运动的影响

Effects of Metformin on Growth and Motility of Melanoma A375 Cells

摘要 目的探讨二甲双脈(Metformin,Met)对黑色素瘤A375细胞生长和运动能力的影响及机制。方法CCK8法检测细胞增殖;流式细胞术检测细胞凋亡率;Transwell检测细胞侵袭;划痕实验检测细胞迁移;蛋白印迹法检测cl-caspase-3、cl-caspase-9、Ki67和MMP-9的蛋白表达,及PI3K/AKT磷酸化;最后,在740Y-P存在情况下检测上述指标。结果与Met 0 mol/L组相比,Met剂量依赖性的降低细胞增殖倍数,迁移细胞数量,划痕闭合率、Ki67、MMP-9,p-PI3K和p-AKT蛋白表达水平,增加细胞凋亡率;且部分逆转740Y-P对上述指标的调节作用。结论二甲双脈抑制黑色素瘤A375细胞生长和运动能力,与调节PI3K/AKT信号通路磷酸化激活有关。 Objective To investigate the effect of metformin ( Met) on the growth and motor ability of melanoma A375 cells and its possible mechanism. Methods CCK8 assay, flow cytometry ,transwell assay, scratch assay and Western blotting were used to detect cell proliferation, apoptosis ,invasion, migration, and the protein expression of cl-caspase-3, cl-caspase-9, Ki67, MMP-9, and PI3K/AKT phosphorylation. Finally, the above indicators were measured in the presence of 740Y-P. Results Compared with the Met 0 mol/L group, Met dose-dependently decreased cell proliferation, the number of invasive cells, wound closure rate, Ki67 , MMP-9, p-PI3K and p-AKT protein expression levels, increased apoptosis rate, and partly reversed the regulatory effects of 740Y-P on the above indicators. Conclusion Metformin could inhibit the growth and motor ability of melanoma A375 cells, and its mechanism is related to the regulation of phosphorylation activation of PI3K/AKT signaling pathway.
