摘要 目的探究hsa-miR-328-3p对人视网膜母细胞瘤WERl-Rb-1生物学活性的影响。方法将hsamiR-328-3pmimic、MMP-16分别或两者共转染于WERI-Rb-1细胞。验证hsa-miR-328-3p与MMP-16的靶向关系,检测各组细胞生长、迁移、侵袭能力及各蛋白表达水平。将对照组及miR-328mimic组WERI-Rb-1细胞分别皮下注射入裸鼠体内,30d后处死、取瘤,检测肿瘤组织各蛋白及基因表达水平。结果MMP-16是hsa-miR-328-3p的靶基因,相比较于pc-MMP-16组,过表达hsa-miR-328-3p能显著抑制MMP-16表达,抑制WERI-Rb-1细胞生长、迁移及侵袭(P<0.01)。与对照组比较,miR-328mimic组肿瘤体降低,hsamiR-328-3p表达量上升(P<0.01)。结论过表达hsa-miR-328-3p可靶向抑制MMP-16表达,从而抑制WERI-Rb-1细胞生长、迁移及侵袭,并降低其上皮间质转化程度。 Objective To explore the effect of hsa-miR-328-3p on the biological activity of human retinoblastoma WERI-Rb-1. Methods hsa-miR-328-3p mimic and MMP-16 were transfected into WERI-Rb-1 cells, respectively, and the two were transfected into WERI-Rb-1 cells. The targeting relationship between hsa-miR-328-3p and MMP-16 was verified. The growth , migration, invasion ability and protein expression levels of each cell group were detected. WERI-Rb-1 cells in the control group and miR-328 mimic group were injected subcutaneously into nude mice. After 30 days, the mice were sacrificed and tumors were obtained. The expression levels of various proteins and genes in tumor tissues were detected. Results MMP-16 is a target gene of hsa-miR-328- 3p. Compared with the pc-MMP-16 group, overexpression of hsa-miR-328-3p significantly inhibited MMP-16 expression and inhibited the growth, migration and invasion of WERI-Rb-1 cells ( P < 0. 01). Compared with the control group, the tumor volume of the miR-328 mimic group decreased ,and the expression of hsaiR-328-3p increased (P < 0. 01). Conclusion Overexpression of hsa-miR-328-3p can inhibit the expression of MMP-16, thereby inhibiting the growth, migration and invasion of WERI-Rb-1 cells and reducing the extent of epithelial mesenchymal transition.
