不同产地连翘EST-SSR标记遗传多样性研究 被引量:3

Genetic Diversity of EST-SSR Markers in Forsythia suspensa(Oleaceae) From Different Habitats

摘要 以采自连翘主产区河南和山西12个产地的77份连翘种质资源为试材,采用EST-SSR分子标记技术分析其遗传多样性,以期探索不同产地连翘的遗传相似性以及其关键影响因素。结果表明:从60对引物中筛选出14对扩增条带清晰,稳定性好的多态性EST-SSR引物,14对引物在77份样品中共扩增出370条带,多态性条带368条,多态位点百分率为99.46%,12个产地的遗传距离范围是0.020 3~0.092 2,平均遗传距离为0.055 8。根据遗传相似系数对12个产地进行聚类分析,在0.935处把12个居群聚为5类;同时对77份样品进行聚类分析,在0.72处把77份样品聚为5类,个体聚类结果与产地聚类结果相似。聚类结果分析表明,连翘的遗传相似性与地理距离没有显著的相关性,主要与生长环境以及海拔等因素有关。试验结果揭示了不同产地连翘遗传相似度的关键因素,为连翘亲缘关系的鉴定、连翘资源的保护与开发奠定了基础。 The genetic diversity of 77 Forsythia suspensa germplasm resources collected from 12 areas of Henan and Shanxi Provinces was analyzed by EST-SSR molecular marker technique,in order to explore the genetic similarity of Forsythia suspensa in different habitats and its key influencing factors.The results showed that 14 pairs of polymorphic EST-SSR primers with clear amplified bands and good stability were screened from 60 pairs of primers.14 pairs of primers amplified 370 bands in 77 samples,including 368 polymorphic bands,and the percentage of polymorphic sites was 99.46%.The genetic distances of the 12 areas ranged from 0.020 3 to 0.092 2,and the average genetic distance was 0.055 8.According to genetic similarity coefficient,12 populations were clustered into 5 groups at 0.935 sites and 77 samples were clustered into 5 classes at 0.935 sites.The result of individual clustering was similar to that of origin.The analysis of clustering results showed that the genetic similarity of Forsythia suspensa had no significant correlation with geographical distance,which was mainly related to growth environment and altitude,etc.The experimental results reveal the key factors of genetic similarity of Forsythia suspensa in different habitats,which lays a foundation for the identification of Forsythia suspensa susceptibility and the protection and development of Forsythia resources.
